Project description:Lung alveolarization is a complex process that involves interactions between several cell types and leads to considerable increase in gas-exchange surface area. The step designated secondary septation includes elastogenesis from interstitial fibroblasts. We used microarrays to detail the global programme of fibroblast gene expression that underly secondary septation, and identified numerous genes that undergo signficant changes in expression Experiment Overall Design: Rat lung fibroblats were extemporaneously isolated at successive stages of postnatal development that flank the septation period for RNA extraction and hybridization on Affymetrix microarrays. Three biological replicates from three different litters were prepared for each time point.

Transcriptome studies of marine natural products 4a and 4d on non-small cell lung cancer A549 cells

Project description:Two novel polyketides, ocellatusones A and B (1 and 2), characterized by either an uncommon oxatricyclo[4.2.1.02,4]nonane skeleton (1) or a mesitylene connected dimethylfuran-3(2H)-one nucleus (2), were isolated in racemic forms from the South China Sea sacoglossan Placobranchus ocellatus, together with seven uncommon polypropionates (3, 4a,4d, 5, and 6). Compounds 1 and 2 were further separated by chiral HPLC to their corresponding enantiomers (±)-1 and (±)-2, respectively. The structures, including absolute configurations, of the new compounds, were unambiguously determined by extensive spectroscopic analysis, quantum chemical computation, and/or X-ray diffraction analysis. The new compounds (+)-1, 4a, 4b, 4d exhibited interesting dose dependent cytotoxic effect on human cancer cell lines, including non-small cell lung cancer and acute promyelocytic leukemia, with IC50 values ranging from 6.1 μM to 28.8 μM. A plausible bio-photosynthetic pathway of these isolates was proposed.

Project description:Rat somatic lung cell had different gene expression level compared with that of the adult cells including rat bone marrow cells(BMC) and rat primary ear fibroblasts(PEF). This difference gave the rat somatic lung cell unique characteristics which could then be compared with the rat iPS cells by genes comparison to show the gene expression difference between rat somatic lung cells and rat iPS cells.
