RNA-seq in Black and White Sesame

Project description:Sesame seeds is an important traditional crop with high oil content and other abundant nutrients which are very beneficial for diet and health of human being. However, the molecular mechanism for metabolite accumulation, especially for oil and phenylpropanoid biosynthesis, is still not very clear in sesame. In this study, the transcriptome profiles of black and white sesame seeds were compared by RNA-sequencing. Transcriptome analysis showed that the expression patterns of genes encoding phenylpropanoid pathway enzymes were different between the two sesame cultivars. Compared with white sesame, most of genes involved in oil biosynthesis were significantly down-regulated in black sesame.

Transcriptional changes in mycorrhizal and non-mycorrhizal soybean roots in presence of the fungal pathogen Macrophomina phaseolina

Project description:Macrophomina phaseolina, a soil borne pathogen with a wide host range, causes the charcoal rot in soybean (Glycine max (L.) Merr). In Argentina, soybean is the most important crop as far as sowing surface cultivation concerns, and for this reason it is important to assess integrated disease management strategies in order to minimize the incidence of this disease and the consequent loss of performance culture. Previous studies have demonstrated the protective effect that arbuscular mycorrhizal can promote on their hosts against various pathogens invasion, especially against soil borne fungi. Therefore, the goal of this study was to analyze the phenomenon of mycorrhizae mediated protection by characterizing transcriptional changes using cDNA microarrays as a tool. The objective of the present study was to investigate, under strict in vitro culture conditions, the global transcriptional change in the roots of pre-mycorrhized soybean plantlets challenged by M. phaseolina as compared to non-mycorrhizal soybean plantlets. The MapMan software was used to distinguish transcriptional change under these conditions, with special emphasis on plant defence response.

Project description:Macrophomina phaseolina is a global devastating necrotrophic fungal pathogen. It causes charcoal rot disease in more than 500 host plants. It is essential to understand the host microbe interaction and the diseases pathogenesis which can ensure global agricultural crop production and security. An array of virulence factors of M. phaseolina were identified which were found to be involved in pathogenesis of other plant pathogenic fungi also. In conclusion the present study has provided a better understanding of how necrotrophic fungi M. phaseolina modulates host plant defensive processes.

Transcription profiling of wheat response to infection by Fusarium pseudograminearum

Project description:Crown rot of wheat, caused by Fusarium pseudograminearum and other Fusarium species is an important disease globally. To understand the host response to challenge by Fp, we examined gene exression changes in the stem base of the wheat variety Kennedy, following inoculation with macroconidia using the Affymetrix GeneChip Wheat Genome Array. Induced genes included mainly those with defensive functions such as genes encoding anti-microbial proteins as well as oxidative stress-related proteins, signalling molecules, and proteins involved in both primary and secondary metabolism. This study is the first comprehensive analysis of the wheat transcriptome during crown rot infection and provides new insights into the host processes involved in plant defence against this pathogen. Experiment Overall Design: There are six samples, three F. pseudograminearum inoculated samples and three mock inoculated samples. Each sample consists of 2cm of stem base from approximately 20 plants.

Project description:The fungal pathogen Fusarium moniliforme causes ear rot in maize. Ear rot in maize is a destructive disease globally caused by Fusarium moniliforme , due to decrease of grain yield and increase of risks in raising livestock by mycotoxins production. Plants have developed various defense pathways to cope with pathogens. We used microarrays to detail the global programme of gene expression during the infection process of Fusarium moniliforme in its host plant to get insights into the defense programs and the host processes potentially involved in plant defense against this pathogen.
