Characterization of the intergenerational impact of in utero and postnatal oxycodone exposure.

Project description:Prescription opioid abuse during and after pregnancy is a rising public health concern. While earlier studies have documented that offspring exposed to opioids in utero have impaired neurodevelopment, a significant knowledge gap remains in comparing the overall development between offspring exposed in utero and postnatally. Adding a layer of complexity is the role of heredity in the overall development of these exposed offspring. To fill in these important knowledge gaps, the current study uses a preclinical rat model mimicking oxycodone (oxy) exposure in utero (IUO) and postnatally (PNO) to investigate comparative and intergenerational effects in the two different treatment groups. While significant phenotypic attributes were observed with the two treatments and across the two generations, RNA sequencing revealed alterations in the expression of key synaptic genes in the two exposed groups in both generations. RNA sequencing and post validation of genes using RT-PCR highlighted the differential expression of several neuropeptides associated with the hypocretin system, a system recently implicated in addiction. Further, behavior studies revealed anxiety-like behaviors and social deficits that persisted even in the subsequent generations in the two treatment groups. To summarize, our study for the first time reveals a new line of investigation on the potential risks associated with oxy use during and after pregnancy, specifically the disruption of neurodevelopment and intergenerational impact on behavior.

MeDIP-seq of ICR mouse sperm with different combinations of in utero undernutrition and postnatal undernutrition

Project description:Maternal caloric restriction during the last week of gestation resulted in low birth weight (LBW) and increased risk of LBW-associated metabolic diseases in adult life. The metabolic phenotypes transmitted to F2 generation by paternal manner without additional altered nutrition. To investigate the mechanism of this intergenerational inheritance, two Cohorts were exposed to different magnitudes of undernutrition both in utero during the last week of gestation and/or postnatal until weaning. We performed MeDIP-seq on the genomic DNA from sperm collected from these mice.

Brain-Derived Extracellular Vesicle microRNA Signatures Associated with In Utero and Postnatal Oxycodone Exposure.

Project description:Oxycodone (oxy) is a semi-synthetic opioid commonly used as a pain medication that is also a widely abused prescription drug. While very limited studies have examined the effect of in utero oxy (IUO) exposure on neurodevelopment, a significant gap in knowledge is the effect of IUO compared with postnatal oxy (PNO) exposure on synaptogenesis-a key process in the formation of synapses during brain development-in the exposed offspring. One relatively unexplored form of cell-cell communication associated with brain development in response to IUO and PNO exposure are extracellular vesicles (EVs). EVs are membrane-bound vesicles that serve as carriers of cargo, such as microRNAs (miRNAs). Using RNA-Seq analysis, we identified distinct brain-derived extracellular vesicle (BDEs) miRNA signatures associated with IUO and PNO exposure, including their gene targets, regulating key functional pathways associated with brain development to be more impacted in the IUO offspring. Further treatment of primary 14-day in vitro (DIV) neurons with IUO BDEs caused a significant reduction in spine density compared to treatment with BDEs from PNO and saline groups. In summary, our studies identified for the first time, key BDE miRNA signatures in IUO- and PNO-exposed offspring, which could impact their brain development as well as synaptic function.

Male rats were sacrified 28 postnatal days and testes were harvested. RNA were extracted with Rneasy mini kit. A pool of RNA from vehicule group was used as the reference group. Each rat submitted to TCDD was compared to reference group. Four biological replicates were processed in a dye swap experimental plan.

Transient postnatal lung changes on alveolar development after in utero and lactation exposure to nicotine

Project description:Purpose: The goal of this study is to address the consequences of in utero and early life nicotine exposure on alveolar phase of lung development in mice Methods: A mouse model of gestational and early life nicotine exposure was used in this study. Briefly, Nicotine (200 mg/L) was supplied immediately after mating and administered to the females in drinking water supplemented with 2% saccharin for the whole period of gestation and lactation (nicotine group). As a control, we used drinking water with 2% saccharin (saccharin group). Pups born from nicotine-exposed or control mothers were collected at different postnatal days (PNDs) and their lungs were subjected to RNAseq, western blot, histological and detailed stereological analysis. Results: Among 13 827 genes tested, nicotine treatment induced an upregulation of 2098 and downregulation of 1859. Pathway analysis performed with MetaCore software (https://portal.genego.com/) (threshold of 2 and p<0.05) identified several different pathways as upregulated upon nicotine exposure, namely cell cycle, epithelial to mesenchymal transition, stem cell related pathway, WNT signaling pathway, stromal-epithelial interaction, cell adhesion and DNA damage. Furthermore, majority of downregulated genes were associated with the immune system. Conclusions: Our results indicate that in utero and early life nicotine exposure induces extensive changes in gene expression following the birth of mice that reflects in accelerated kinetics of lung development. This effect, however, is transient, and not lomger detectable by pnd16. On the other hand, nicotine decreased proliferation and endothelial cell density detected at pnd16, thereby affecting on of two main cell lines involved in oxygen exchange. This results are important in the context of growing interest for nicotine patch replacement therapy and electronic cigarettes.
