The hyperlipemia protocol was as follows:mice post natal 4 weeks fed normal diet or subjet to 60% high fat diet for 23 weeks, the mice were immediately sacrificed for liver and plasma collection.

RNA-next generation sequencing of intestine tissue from mice fed high fat diet or/and sugar drink

Project description:Purpose: NGS was used to determine if a distinct transcriptomic profile is observed among the experimental mice fed four different dietary components. Methods: We carried out RNA-Seq analysis of ileum tissue from 6 weeks male mice ad libitum fed for 10 weeks a high liquid sugar (23% (w/v)) or/and high fat (60% Kcal from fat) diet. The combined effect of sugar drink and high fat diet (HF-Sugar) was compared with sugar drink only (NF-Sugar), or high fat diet only (HF), or control diet that was plain water and normal fat diet (NF). Results: RNA-Seq revealed sample-specific clusters that included genes responding to each experimental diet. We found only addition of sugar drink to high fat group (HF-Sugar) not NF-Sugar and HF, there was a significant enrichment in biological functions relating to Inflammatory/Immune Responses, especially including dendritic cell (DC) and T cell related signaling pathway. Conclusions: Taken together, our data demonstrate that sugar drink synergistically promotes and exacerbates inflammatory responses driven by the high fat diet.

Livers of mice fed an atherogenic diet for 6 or 24 weeks

Project description:To address the molecular basis for atherogenic diet-induced liver injury, we performed microarray analysis using livers at early (6 weeks) and pre-cirrhosis stages (24 weeks) in the development of steatohepatitis. The Atherogenic diet up-regulated the gene expression for fatty acid synthesis, inflammatory cytokines, oxidative stress, and fibrosis, and with down-regulation of fatty acid beta-oxidation. The addition of a high-fat component to the Atherogenic diet up-regulated the gene expression for fatty acid synthesis and transport pathways and down-regulated some of the antioxidant genes. Project description:We investigated remodeling of the mitochondrial proteome to determine mechanisms of changes to lipid oxidation following high-fat feeding. C57BL/6J mice consumed either a high-fat diet (HFD, 60% fat) or low fat diet (LFD, 10% fat) for 12 weeks. Mice were fasted 4 hours then anaesthetized by sodium pentobarbital for tissue collection. A mitochondrial-enriched fraction was prepared from gastrocnemius muscles and underwent proteomic analysis by high-resolution mass spectrometry.

Expression of obese mice subjected to diet induced weight loss for 60 days

Project description:We used microarrays to study the gene expression dynamics during caloric restricted weight loss in obese mice after feeded with high fat diet. C57BL/6J male mice were placed on a high fat diet at 10-11 weeks of age in individual cages and kept on the diet until they reached ~ 41 g. After all the baseline (41g) measurement were made, each mouse was fed 75% of their ad libitum food intake in two divided daily "meals". The high fat diet was maintained.

Maternal dietary vitamin A deficiency and diet dependent liver gene expression in mice

Project description:Initiation of a vitamin A deficient diet in mid-gestation, maintained in the post-weaning diet is sufficient to cause liver and serum retinoid depletion. Wild type C57Bl/6J timed mated pregnant dams were administered either a defined vitamin A sufficient low fat (12 percent kcal from fat) diet or matched vitamin A deficient diet from embryonic day 10.5. Vitamin A sufficient offspring were weaned onto either a high fat diet (60 percent kcal from fat) or maintained on the low fat 12 percent kcal from fat diet for 11 weeks (14 weeks of age). Gestational vitamin A deficient offspring were maintained on the same vitamin A deficient diet until 14 weeks of age. The impact of the maternal diet on a post-weaning high fat diet was compared to a standard maternal breeder diet followed by the post-weaning high fat diet.
