Project description:Alloimmune T cell mediated gastrointestinal graft-versus-host disease (GI-GVHD) occur after bone marrow translantation (BMT). GI-GVHD cause significant morbidity and mortality. Immunosuppression therapies have shown good results with adverse effects, but are still incomplete. The biology of GVHD are often understood from the immune cell perspectve, but the pathogenesis and severity from host IEC traget cell perspective remain undefined. We used next generation sequencing transcriptome to detail the gene expression of colonic epithelial cells underlying the profile changes between the syngeneic recipient and allogeneic recipient.

Haploidentical Donor Bone Marrow Transplant in Treating Patients With High-Risk Hematologic Cancer

Project description:This phase II trial studies how well giving fludarabine phosphate, cyclophosphamide, tacrolimus, mycophenolate mofetil and total-body irradiation together with a donor bone marrow transplant works in treating patients with high-risk hematologic cancer. Giving low doses of chemotherapy, such as fludarabine phosphate and cyclophosphamide, and total-body irradiation before a donor bone marrow transplant helps stop the growth of cancer cells by stopping them from dividing or killing them. Giving cyclophosphamide after transplant may also stop the patient’s immune system from rejecting the donor’s bone marrow stem cells. The donated stem cells may replace the patient’s immune system cells and help destroy any remaining cancer cells (graft-versus-tumor effect). Sometimes the transplanted cells from a donor can also make an immune response against the body’s normal cells. Giving tacrolimus and mycophenolate mofetil after the transplant may stop this from happening

Project description:We found that mesenchymal stem cells isolated from colonic tissue express significantly increased expression of Ptgs2 or cox-2 compared to that of mesenchymal stem cells isolated from the bone marrow, one of the most common tissues from which this cell type is derived. We hypothesized that the localization of these cells in the colon exposes them to specific environmental factors that induce and/or maintain increased constitutive levels of cox-2 uniquely in this cell type. In order to identify possible candidate molecules regulating cox-2 in these cells, we decided to compare the transcriptomes of colonic mesenchymal stem cells and bone marrow-derived mesenchymal stem cells. Mesenchymal stem cells were isolated by fast plastic adherence after collagenase type I-mediated dissociation of the tissue of origin. Cells were maintained in culture in low glucose DMEM with 10mM HEPES and 10% FBS. Cells were passaged at 90-100% confluency. Approximately 4 million cells from four MSC lines, two from colon and two from bone marrow, were lysed between passages 5 and 10. Total RNA was isolated, amplified, and hybridized to Affymetrix MOE230.2 chips.

HSC colony many years post allogeneic bone marrow transplant TGS

Project description:This study looks at human donors and recipients of allogeneic blood & marrow marrow transplantation. Each transplant "pair" (donor & recipient) underwent the transplant procedure 10 -30 years prior to sampling - the majority for an underlying diagnosis of leukemia. We will perform whole-genome sequencing on tens to hundreds of single HSPC (haematopoietic stem and progenitor cell)-derived colonies per individual. We will infer the phylogeny of these cells (those from both donor & recipient within a pair) by analysing the somatically acquired mutations within each cell. The structure of the phylogeny will allow estimates of hitherto unknown parameters such as the number of HSCs that engraft long-term, the dynamics of the engraftment process on the HSC level, and the additional mutation burden acquired through the arduous transplant procedure. This TGS part of the study is used for assessing the contribution of clones to mature blood subsets, and better timing mutations relative to transplantation.
