Project description:To identify the impact of ATF4 deletion on the expression of collagen-associated and fibroblast activation genes. We used microarrays to detail the gene expression profile on lung fibroblasts post ATF4 deletion. Many genes to be differentially expressed, with a profound reduction in expression of collagen-associated and fibroblast activation genes in LFBD/D. We identified that ECM organization/degradation and collagen biosynthesis pathways to be the most impaired in LFBD/D.

Project description:Intratracheal transfer of isolated lung fibroblasts in bleomycin-induced lung fibrosis recapitulates the activation process of lung fibroblasts after epithelial injury. In order to investigate gene expression signatures of transferred fibroblasts, we purified transferred fibroblasts 2, 4, and 7 days after the transfer and performed transcriptome analysis. We also isolated Acta2 high and low cells by using Acta2-mKO1 reporter mice 4 days after the transfer.
