We performed the single cell transcriptomic analysis of PBMC from rheumatoid arthritis (RA) patient, and monocyte populations were extracted during the analysis. CD127 expression associated expression pattern of inflammatory genes was identified in RA patient's blood monocytes.

To characterize RA at the molecular level and to uncover key pathomechanisms, we performed whole-genome gene expression analyses. Within this study, the proteomes of serum obtained from rheumatoid arthritis patients, and appropriate human controls, were analysed using liquid chromatography-tandem mass spectrometry. Project description:We compared whole CD4+ and CD8+ T cells from frozen PBMC samples that were collected before and after treatment initiation of each patient with rheumatoid arthritis. Lists consisting of 858 and 950 differentially expressed genes were created from CD4 and CD8, respectively, and these were used for enrichment analysis. As a result, we found that certain pathways were downregulated after TCZ treatment in both CD4+ and CD8+ T cells, including mechanistic target of rapamycin complex 1 (mTORC1) signaling, the IL-2 pathway, and IFN-related genes.

Project description:Background: In rheumatoid arthritis (RA), only a subset of patients develop an irreversible bone destruction. Our aim was to develop a microRNA (miR)-based osteoclast-related signature to predict erosiveness in RA. Results: We identified 5 miRs from PBMC-derived osteoclasts differentially expressed in RNASeq also in qPCR assessment. Conclusion: We found 5 miRNAs differentially refulated in erosive rheumtoid arhtritis compare to no erosve patient. This study could improve the current approach to identify RA subjects at risk of erosions, by adding biomarker signatures based on the profile of miRs in the osteoclasts.
