Project description:Primary objectives: Taux de contrôle de la maladie à 12 mois (12 mois après randomisation) Primary endpoints: La Durée de Contrôle de la Maladie (DCM) est définie comme l’intervalle de temps entre la date de randomisation et la date de progression : - comparé à l’évaluation de l’inclusion si progression au cours de la première séquence thérapeutique - comparé à l’évaluation avant la dernière reprise de la chimiothérapie au cours d’une séquence de traitement par chimiothérapie - ou au décès (dû à la progression)Une progression au cours d’un ILC n’est pas prise en compte pour le calcul de la DCM. En revanche, chez les patients ayant définitivement arrêté la chimiothérapie pour intolérance, la progression sous anti-angiogénique seul sera prise en compte. Les patients vivants sans progression ou décédés pour une autre raison que la progression seront censurés à la date de dernière évaluation ou du décès. La progression est définie selon les critères RECIST. Une ascension isolée des marqueurs tumoraux n’est pas considérée comme une progression.

Project description:Primary objectives: Parte 1: SAFETY RUN-IN PART Determinar la Dosis Máxima Tolerada (DMT) y la Dosis Recomendada de Fase II (DRF2) de MSC1936369B combinado con FOLFIRI como tratamiento de segunda línea en pacientes con Cáncer colorrectal metastásico con K-Ras mutado (CCRm). Parte 2: PARTE DE ASIGNACIÓN ALEATORIZADA FASE II Valorar la actividad anti-tumoral de MSC1936369B combinado con FOLFIRI como tratamiento de segunda línea en pacientes con CCRm K-Ras mutado en términos de Supervivencia libre de progresión (SLP). Primary endpoints: Safety Run-in Part Toxicidad limitante de dosis (TLD) utilizando los Criterios Comunes de Terminología NCI para Acontecimientos Adversos v3.0 (CTCAE), evaluada durante el primer ciclo de tratamiento. Se utilizará el número y la incidencia de TLDs por nivel de dosis en el análisis estadístico primario de la Safety Run-in Part. Parte de Asignación Aleatorizada Fase II Tiempo de supervivencia libre de progresión (SLP), definido como el tiempo (en meses) desde la fecha de asignación aleatorizada hasta la primera progresión de la enfermedad según lo notificado y documentado por el investigador (ej., progresión radiológica por RECIST v1.0) o muerte por cualquier causa. Se censurará la SLP en el momento de posterior tratamiento antes de la progresión. En el análisis estadístico principal de la Parte de Asignación Aleatorizada Fase II se utilizará la Razón de Riesgo (RR) de la SLP del grupo experimental versus el grupo control.

Project description:Understanding and quantifying the effects of environmental factors influencing the variation of abundance and diversity of microbial communities was a key theme of ecology. For microbial communities, there were two factors proposed in explaining the variation in current theory, which were contemporary environmental heterogeneity and historical events. Here, we report a study to profile soil microbial structure, which infers functional roles of microbial communities, along the latitudinal gradient from the north to the south in China mainland, aiming to explore potential microbial responses to external condition, especially for global climate changes via a strategy of space-for-time substitution. Using a microarray-based metagenomics tool named GeoChip 5.0, we showed that microbial communities were distinct for most but not all of the sites. Using substantial statistical analyses, exploring the dominant factor in influencing the soil microbial communities along the latitudinal gradient. Substantial variations were apparent in nutrient cycling genes, but they were in line with the functional roles of these genes. 300 samples were collected from 30 sites along the latitudinal gradient, with 10 replicates in every site
