Project description:To further explore the differential expression profile of super-enhancer long noncoding RNA (LncRNA) in human bladder cancer, we have employed super-enhancer lncRNA microarray expression profiling as a discovery platform to identify potential differential expression profile of super-enhancer lncRNA between human bladder cancer cell (UM-UC-3 cell) and urothelial immortalized cell (SV-HUC-1 cell). Results showed that a large number of differentially expressed super-enhancer lncRNA were found between UM-UC-3 cell and SV-HUC-1 cell. In this study, We verified 5 up-regulated differentially expressed super-enhancer lncRNAs using qPCR, of which the highest fold change is LINC00162. Then we further explored the biological function and mechanism of LINC00162 in bladder cancer in this study.

Project description:To further explore the differential expression profile of circRNA in human bladder cancer, we have employed circRNA microarray expression profiling as a discovery platform to identify potential differential expression profile of circRNA between human bladder cancer cell (UM-UC-3 cell) and urothelial immortalized cell (SV-HUC-1 cell). Results showed a large number of differentially expressed circRNAs. In this study, we screened the eight circRNAs that were most significantly downregulated in the circRNA differential expression profile of bladder cancer cells, designed primers across the splice site, and detected them using qPCR. We found that hsa_circ_0047253 (circLAMA3) was most significantly decreased in the bladder cancer cells, then its biological function and action mechanism have been further explored in this study.

A conditionally immortalized human pancreatic beta cell line

Project description:Access to an unlimited number of human pancreatic beta cells represents a major challenge in the field of diabetes to better dissect human beta cell functions and to make significant progress in drug discovery and cell replacement therapies. We previously reported the generation of the EndoC-bH1 human beta cell line that was generated by targeted oncogenesis in human fetal pancreases followed by in vivo cell differentiation in mice. Such cell line displayed many functional properties of adult beta cells. Here we devised a novel strategy to generate conditionally immortalized human beta cell lines based on CRE-mediated excision of immortalizing transgenes. The resulting EndoC-bH2 cell line can be massively amplified in vitro. Transgenes are next efficiently excised upon CRE expression leading to cell proliferation arrest and strong enhancement of beta cell specific features such as insulin expression, content and secretion. Excised EndoC-bH2 cells are close to authentic human beta cells and represent a unique tool to further study beta cell function and to understand why adult human beta cells are refractory to proliferation and how to achieve drug-dependent mobilization towards beta cell expansion.

Generation and Characterization of an immortalized human mesenchymal stromal cell line model [Affymetrix]

Project description:Human mesenchymal stromal cells (hMSCs) show great potential for clinical and experimental use. Self-renewal and multipotent differentiation capacity are key elements in the increasing interest of this cell type. However, a disadvantage of primary hMSCs is the limited in vitro lifespan, but this can be overcome by introduction of the catalytic subunit of human telomerase reverse transcriptase (TERT). In this paper, the generation and characterization of a TERT-immortalized, non-tumorigenic human bone marrow-derived stromal mesenchymal model cell line is described. The resulting cell line, iMSC#3, maintained capacity to differentiate into osteoblasts and adipocytes, and growth characteristics comparable to primary hMSCs into after long-term culturing (155 population doublings). A detailed characterization of the mRNA- and microRNA transcriptomes during adipocyte differentiation also showed that the iMSC#3 recapitulates this process at the molecular level. Furthermore, iMSC#3 showed no major differences from hMSCs regarding surface marker expression. The cell line had a normal karyotype, and high-resolution array comparative genomic hybridization confirmed that the cells in general had normal copy number. The gene expression profiles of immortalized and primary hMSCs were also similar, whereas the corresponding methylation profiles were more diverse. In conclusion, a human bone marrow-derived stromal cell line was developed, which can be used for basic studies of mesenchymal cells and to functionally study transgenes that might play a role in oncogenic transformation.
