Project description:We examined the transcriptome induced by GmNAC81 overexpression and leaf senescence and showed that GmNAC81 further modulates leaf senescence by regulating an extensive repertoire of functionally characterized senescence-associated genes (SAGs). GmNAC81 overexpression also uncovered the regulation of typical drought-responsive genes. Key regulators and effectors of ABA signaling were suppressed by GmNAC81 overexpression.

Bikaneri Nerma was used for the gene expression analysis. Cotton plants were subjected to drought stress at peak flowering stage. Leaf samples were collected when the soil moisture content was 19.5% which is 50% of the normal control plots. Gene expression profiles in drought induced and their respective control samples were analyzed using Affymertix cotton Genechip Genome arrays to study the global changes in the expression of genome.

Project description:Kappaphycus alvarezii seaweed extract (KSWE) has been known for its plant biostimulant and stress alleviation activities on various crops. However, very few reports are available depicting its impact at the molecular level, which is crucial in identifying the mechanism of action of KSWE on plants. Here, maize leaf tissue of control and KSWE treated plants were analyzed for their transcriptional changes under drought stress. KSWE was applied foliarly at the V5 stage of maize crop under drought and leaf transcriptome analysis was performed. It was found that total of 380 and 631 genes were up- and down- regulated, respectively due to the application of KSWE. Genes involved in nitrate transportation, signal transmission, photosynthesis, transmembrane transport of various ions, glycogen and starch biosynthetic processes were found up-regulated in KSWE treated plants. While genes involved in the catabolism of polysaccharide molecules such as starch as well as cell wall macromolecules like chitin and protein degradation were found down-regulated. An overview of differentially expressed genes involved in metabolic as well as regulatory processes in KSWE treated plant was also analyzed via Mapman tool. Phytohormone signaling genes such as cytokinin-independent 1 (involved in cytokine signal transduction), Ent-kaurene synthase and GA20 oxidase (involved in gibberellin synthesis) and gene of 2-oxoglutarate-dependent dioxygenase enzyme activity (involved in ethylene synthesis) were found up-regulated while 9-cis-epoxycarotenoid dioxygenase (a gene involved in abscisic acid synthesis) was found down regulated due to the application of KSWE. Modulation of gene expression in maize leaf tissue in response to KSWE treatment elucidate mechanisms to ward off drought stress which can be extended to understand similar phenomenon in other crops as well. . This molecular knowledge can be utilized to make the use of KSWE more efficient and sustainable.

Project description:Switchgrass (Panicum virgatum) is a perennial crop producing deep roots thus highly tolerant to soil water deficit conditions. However, seedling establishment in field is very susceptible to prolonged and periodic drought stress. In this study, a “sandwich” system simulating a gradual water deletion process was developed. Switchgrass seedlings were subjected to a 20-day gradual drought treatment process when soil water tension was increased to 0.05 MPa (moderate drought stress) and leaf physiological properties had expressed significant alteration. Drought-induced changes in leaf proteomes were identified using the relative and absolute quantitation (iTRAQ) labeling method followed by nano-scale liquid chromatography mass spectrometry (nano-LC-MS/MS) analysis. Additionally, total leaf proteins were processed using a combinatorial library of peptide ligands to enrich for lower abundance proteins. Both total proteins and those enriched samples were analyzed to increase the coverage of the quantitative proteomics analysis. A total of 7,006 leaf proteins were identified, and 257 (4% of the leaf proteome) expressed a significant difference (P < 0.05, fold change < 0.6 or > 1.7) from the non-treated control to drought-treated conditions. Results from this study, in addition to revealing molecular responses to drought stress, provide a large number of proteins (candidate genes) that can be employed to improve switchgrass seedling growth and establishment under soil drought condition.

Project description:We use the gowth zone of the maize leaf as a model system to study the growth reduction in response to drought stress. The spatial gradient and the relatively large size of the maize leaf allowed us to sample at a subzonal rezolution and to examine different developmental stages at the same time. We compared the response to different levels of drought stress (mild and severe) of proliferating (meristem), expanding (elongation zone) and differentiated (mature zone) tissue.

Project description:To explore the usefulness of Brachypodium distachyon for drought studies, a reproducible in soil drought assay was developed. Spontaneous soil drying led to a 45% reduction in leaf size, and this most mostly due to a decrease in cell expansion, whereas cell division remained largely unaffected by drought. To investigate the molecular basis of the observed leaf growth reduction, Brachypodium leaf 3 was dissected in three zones, namely the proliferation, expansion and mature zone, and subjected to transcriptome analysis using a Affymetrix whole-genome tiling array. This approach allowed us to highlight that transcriptome profiles of different developmental leaf zones respond differently to drought. Several genes and biological processes involved in drought tolerance were identified. Mainly, we observed an increased energy availability in the proliferation zone along with an upregulation of sterol synthesis that may influence membrane fluidity. 24 samples of Brachypodium leaf 3 harvested about 24 hours after the emergence of leaf 3, mild drought stress was performed for 2 biological replicates, severe drought stress and control condition was performed for 3 biological replicates. 3 developmental zones, namely the proliferation, expansion and mature zone, are dissected from leaf 3.

Project description:This study was aimed at deciphering the impact of drought and heat on genome-wide gene expression in flag leaf of barley. We employed high-throughput sequencing of mRNA to identify genes that are associated with response to drought or heat and to their combination. Our study demonstrated that under combined stress, drought was the dominant factor affecting genes expression. It was also confirmed for phenotypic traits and chlorophyll fluorescence parameters. Drought- and heat-responsive genes were associated majorly with photosynthesis, abscisic acid signaling and lipids transport. Dehydrin encoding genes were found to be universal stress-responsive genes. Stress-induced genes specific to the flag leaf size were also found. This research provided novel insight into molecular mechanisms of barley flag leaf that determine drought and heat response, also during their co-occurrence.

Project description:The effect and responses to drought stress have been analyzed in Quercus ilex seedlings by using a non-targeted metabolomic approach, implementing previous studies pub-lished in which other -omics platforms, transcriptomics, and proteomics, have been em-ployed. This work is aimed at characterizing the Q. ilex leaf metabolome, determining possible mechanisms and molecular markers of drought tolerance, and identifying puta-tive bioactive compounds. Six-month-old seedling leaves, subjected to drought stress im-posed by water withholding under high temperature and irradiance conditions, were col-lected when leaf fluorescence decreased by 20 % (day 17) and 45 % (day 24) relative to ir-rigated seedlings. A total of 3934 compounds were resolved, with 616 being variable, and 342 identified, belonging to five chemical families. Out of the identified compounds 33 were variable, mostly corresponding to amino acids, carboxylic acids, benzenoids, flavo-noids and isoprenoids. Epigallocatechin, ellagic acid, pulegone, indole-3-acrylic acid and dihydrozeatin-O-glucoside were up-accumulated under drought conditions at both sam-pling times. An integrated multi-omics analysis of phenolic compounds and related en-zymes was performed, revealing that some enzymes involved in the flavonoid pathways (chalcone synthase, anthocyanidin synthase and anthocyanidin reductase) were up-accumulated at day 24 in non-irrigated seedlings. Some putative markers of drought tol-erance to drought in Q. ilex are proposed for assisting breeding programs based on the se-lection of elite genotypes.

Project description:Results: In this study, the ears at the V9 stage, kernels and ear leaf at the 5DAP (days after pollination) stage of maize were used for morphological, physiological and comparative transcriptomics analysis to understand the different features of “sink” or “source” organs and the effects on kernel yield under drought stress conditions. The ABA-, NAC-mediate signaling pathway, osmotic protective substance synthesis and protein folding response were identified as common drought stress response in the three organs. Tissue-specific drought stress responses and the regulators were identified, they were highly correlated with growth, physiological adaptation and yield loss under drought stress. For ears, drought stress inhibited ear elongation, led to the abnormal differentiation of the paired spikelet, and auxin signaling involved in the regulation of cell division and growth and primordium development changes. In the kernels, reduced kernel size caused by drought stress was observed, and the obvious differences of auxin, BR and cytokine signaling transduction appeared, which indicated the modification in carbohydrate metabolism, cell differentiation and growth retardation. For the ear leaf, dramatically and synergistically reduced the expression of photosynthesis genes were observed when suffered from drought stress, the ABA- and NAC- mediate signaling pathway played important roles in the regulation of photosynthesis. Conclusions: Transcriptomic changes caused by drought were highly correlated with developmental and physiological adaptation, which was closely related to the final yield of maize, and a sketch of tissue- and developmental stage-specific responses to drought stress in maize was drafted.
