Project description:Despite intensive studies that improved our understanding of the disease mechanisms and multiple clinical trials, amyotrophic lateral sclerosis (ALS) remains an uncurable disease. Riluzole and Edaravone, two currently approved drugs, provide limited quality-of-life improvements in ALS patients. This emphasises the importance of searching for new drugs and therapeutic approaches that might be used, most probably in combination, for efficient treatment of ALS. A neuroprotective ability of certain gamma-carbolines, including Dimebon/Latrepirdine and its derivates, has been demonstrated in several models of neurodegeneration, suggesting that these compounds have a potential as components of a multiplex disease-modifying therapy of ALS. In this study we showed that a chronic treatment with a novel Dimebon bioisostere, DF402, delays the onset and slows the progression of pathology in the transgenic model of motor neuron disease caused by expression of pathogenic truncated form of human FUS protein. By using CatWalk gait analysis we have revealed parameters of transgenic animal gait that display changes before manifestation of clinical signs of pathology and parameters that are changed at the pre-symptomatic stage in response to DF402 treatment. RNAseq was used to compare the spinal cord transcriptomes of wild type, untreated and DF402-treated FUS transgenic mice. We found that at the early pre-symptomatic stage a limited number of genes significantly change expression in the spinal cord of transgenic mice and demonstrated that for 60% of these genes DF402 treatment causes the reversion of expression towards levels typical for expression of these genes in the spinal cord of wild type animals.

Project description:AimsTo assess effects of DF402, a bioisostere of Dimebon/Latrepirdine, on the disease progression in the transgenic model of amyotrophic lateral sclerosis (ALS) caused by expression of pathogenic truncated form of human FUS protein.MethodsMice received DF402 from the age of 42 days and the onset of clinical signs, the disease duration and animal lifespan were monitored for experimental and control animals, and multiple parameters of their gait were assessed throughout the pre-symptomatic stage using CatWalk system followed by a bioinformatic analysis. RNA-seq was used to compare the spinal cord transcriptomes of wild-type, untreated, and DF402-treated FUS transgenic mice.ResultsDF402 delays the onset and slows the progression of pathology. We developed a CatWalk analysis protocol that allows detection of gait changes in FUS transgenic mice and the effect of DF402 on their gait already at early pre-symptomatic stage. At this stage, a limited number of genes significantly change expression in transgenic mice and for 60% of these genes, DF402 treatment causes the reversion of the expression pattern.ConclusionDF402 slows down the disease progression in the mouse model of ALS, which is consistent with previously reported neuroprotective properties of Dimebon and its other bioisosteres. These results suggest that these structures can be considered as lead compounds for further optimization to obtain novel medicines that might be used as components of complex ALS therapy.

Project description:A number of mutations in a gene encoding RNA-binding protein FUS was linked to the development of a familial form of amyotrophic lateral sclerosis known a FUS-ALS. C-terminal truncations of FUS by a nonsense or frameshift mutations lead to the development of FUS-ALS with a particularly early onset and fast progression. However, even in patients with this aggressive form of the disease the function of motor neurons is not noticeably compromised for at least a couple of decades suggesting that until accumulation in the cytoplasm of pathogenic FUS lacking its C-terminal nuclear localisation signal does not reach a critical threshold, motor neurons are able to tolerate its permanent production. In order to identify how the nervous system responds to low levels of pathogenic variants of FUS we produced and characterised a mouse line, L-FUS[1-359], with a low level of neuronal expression of a highly aggregation prone and pathogenic form of C-terminally truncated FUS. In contrast to mice with substantially higher level of expression of the same FUS variant that develop severe early onset motor neuron pathology, L-FUS[1-359] mice do not develop any sign of pathology even at old age. Nevertheless, we detected substantial changes in the spinal cord transcriptome of these mice comparing to the wild type littermates. We suggest that at least some of these changes reflect activation of cellular mechanisms compensating to potentially damaging effect of pathogenic FUS production. Further studies of these mechanism might reveal effective target for therapy of FUS-ALS and possibly, other forms of ALS

Project description:Frontotemporal dementia (FTD) is an uncommon cause of behavioural change in adults under the age of 50. A 44-year-old man presented with progressive neuropsychiatric disturbance characterised by social withdrawal, apathy, loss of empathy, motor stereotypies and hyperorality. Cognitive testing identified severe impairment, including executive dysfunction. MR scan of the brain showed bilateral symmetrical frontal atrophy. There was no relevant family history, and targeted genetic testing for FTD-associated variants in MAPT, GRN and C9orf72 genes proved negative. He became more withdrawn with disinhibited behaviour; his condition progressively worsened and he died 6 years later. The pathological diagnosis was frontotemporal lobar degeneration with fused-in-sarcoma (FUS) pathology, a rare sporadic cause of FTD, accounting for only 5%-10% of cases, its characteristic features including very young onset, motor stereotypies and hyperorality.

FUS regulates nuclear RNA, but its role at the synapse is poorly understood. Here, we used super-resolution imaging to determine the physiological localization of extranuclear, neuronal FUS and found it predominantly near the vesicle reserve pool of presynaptic sites. Using CLIP-seq on synaptoneurosome preparations, we identified synaptic RNA targets of FUS that are associated with synapse organization and plasticity. Synaptic FUS was significantly increased in a knock-in mouse model of ALS-FUS, at presymptomatic stages. Despite apparently unaltered synaptic organization, RNA-seq of synaptoneurosomes highlighted age-dependent dysregulation of glutamatergic and GABAergic synapses. Our study indicates that FUS relocalization to the synapse in early stages of ALS-FUS results in synaptic impairment, potentially representing an initial trigger of neurodegeneration.

Project description:The aim of our study is to identify the role of FUS in shaping the transcriptome. RNA-seq of two FUS KO clones was performed and compared to wt; for each, four replicates were sequenced. RNA molecules associated with the FUS protein were determined by means of a RNA immuno-precipitation, followed by high-throughput sequencing. Total RNA was used as a control. SH-SY5Y cells were used for both experiments. RNA-seq: 4 wt samples, 4 A4 KO samples, 4 A5 KO samples. RIP-seq: 1 input control sample, 3 anti-FUS IP replicates.

Project description:Unraveling the mechanistic link connecting the multiple RNA-binding proteins (RBPs) associated with amyotrophic lateral sclerosis (ALS) is critical for identifying novel therapeutics. Mutations in the RBP FUS cause the third most common genetic form of ALS. Here, we show that motor neurons (MNs) of FUS-ALS patients manifest heterogeneous levels of cytoplasmic FUS. Using neurons differentiated from induced pluripotent stem cells (iPSCs) carrying a FUS-eGFP reporter, we demonstrate that pronounced cytoplasmic FUS mislocalization is linked to aberrant protein degradation. We also show that the P525L FUS mutation reduces the interaction of FUS with RBPs, including hnRNPA1, hnRNPA2B1, EWSR1, and TAF15, facilitating FUS aggregation. Additionally, RBP levels are decreased, inducing neurodegeneration. We use patient spinal cord to demonstrate that MNs containing aggregated FUS have reduced RBP content compared to MNs lacking FUS aggregates. Finally, we demonstrate that small molecules inducing autophagy, including PQR309, a brain penetrant compound in clinical trials, restore proteostasis.
