Project description:COVID-19 patients often develop severe cardiovascular complications, but it remains unclear if these are caused directly by viral infection or are secondary to a systemic response. Here, we examine the cardiac tropism of SARS-CoV-2 in human pluripotent stem cell-derived cardiomyocytes (hPSC-CMs) and smooth muscle cells (hPSC-SMCs). We find that that SARS-CoV-2 selectively infects hPSC-CMs through the viral receptor ACE2, whereas in hPSC-SMCs there is minimal viral entry or replication. After entry into cardiomyocytes, SARS-CoV-2 is assembled in lysosome-like vesicles and egresses via bulk exocytosis. The viral transcripts become a large fraction of cellular mRNA while host gene expression shifts from oxidative to glycolytic metabolism and upregulates chromatin modification and RNA splicing pathways. Most importantly, viral infection of hPSC-CMs progressively impairs both their electrophysiological and contractile function, and causes widespread cell death. These data support the hypothesis that COVID-19-related cardiac symptoms can result from a direct cardiotoxic effect of SARS-CoV-2.

Project description:Pluripotent stem cell (PSC)-derived electrically excitable cells provide a unique window into human disease and development, but they remain electrically immature compared to their in vivo counterparts, partially due to the lack of chronic stimulation. New classes of on-demand, electrically active biomaterials are being employed to enhance cell functions, and here, we fabricated biocompatible, electrospun polymer nanofibers containing light-reactive reduced graphene oxide (rGO). Fiber size, stiffness, and electrical conductivity scaled with rGO concentration which in turn produced conduction-dependent effects in PSC-derived cardiomyocytes and neurons; with acute light stimulation on scaffolds containing rGO, cardiomyocytes exhibited more calcium handling activity and were more synchronous, and neurons showed more peaks with higher frequency. Chronic repetitive, daily light stimulation of nanofibers integrated into mature brain organoids caused organoids to become increasingly more electrically responsive to stimulation over time, mature synapses of excitable neurons, and activate photoreceptor lineage pathways. This work outlines a tunable method where the electrical function of electrically excitable, PSC-derived cells can be titrated with rGO fibers and light stimulation, and it suggests that repetitive light stimulation may provide a novel method for retinal cell differentiation in organoids.

Electrical Stimulation Mitigates Muscle Degradation Shift in Gene Expressions during 12-hour Mechanical Ventilation

Project description:Ventilator-induced diaphragm dysfunction (VIDD), a dysfunction of the diaphragm muscle caused by prolonged mechanical ventilation (MV), is an important factor that hinders successful weaning from ventilation. We evaluated the effects of electrical stimulation of the diaphragm muscle (pulsed current with off-time intervals) on genetic changes during 12 hours of MV (E-V12). Rats were divided into four groups: control, 12-hour MV, sham operation, and E-V12 groups. Transcriptome analysis using an RNA microarray revealed that 12-hour MV caused upregulation of genes promoting muscle atrophy and downregulation of genes facilitating muscle synthesis, suggesting that 12-hour MV is a reasonable method for establishing a VIDD rat model. Of the genes upregulated by 12-hour MV, 18 genes were not affected by the sham operation but were downregulated by E-V12. These included genes related to catabolic processes, inflammatory cytokines, and skeletal muscle homeostasis. Of the genes downregulated by 12-hour MV, 6 genes were not affected by the sham operation but were upregulated by E-V12. These included genes related to oxygen transport and mitochondrial respiration. These results suggested that 12-hour MV shifted gene expression in the diaphragm muscle toward muscle degradation and that electrical stimulation counteracted this shift by suppressing catabolic processes and increasing mitochondrial respiration.
