Project description:Ingestion of a diet with a restricted Methionine (MR) content results in robust activation of hepatic stress pathways regulated by PERK, ATF4, and Nfe2l2. In addition, genes regulating hepatic lipogenesis are downregulated. So far, data were obtained using a mix of elemental amino acids as the dietary protein source, which was replaced in this experiment with casein. To reduce methionine content, casein was oxidized and methionine was added back to a concentration similar to the diets using elemental amino acids as protein source. Results show that mice reduce body weight and adiposity on MR containing oxidized casein similar to MR containing elemental amino acids. Similarly, energy expenditure, as well as food and water intake were increased. The RNAseq experiment was conducted to compare the effect of MR using these two protein sources on the hepatic transcriptome.

The effect of dietary methionine restriction using different protein sources on the transcriptome of mouse livers

Project description:Dietary Methionine restriction (MR) has been demonstrated to induce a set of hepatic responses, including activation of hepatic stress response and suppression of hepatic lipogenesis. In the current study, we used two different protein sources to achieve MR. Casein was used in normal protein (NP, 20% casein) and low protein (LP, 5% casein) diet. Soy protein was used in 20Soy (20% soy), 20Soy+SAA (20% soy, Met and Cys was added back to match their concentration in NP diet), and 10Soy (11.5% soy, the Met concentration matches LP diet). Results show that mice that were fed LP and 10Soy diet have lower body weight and adiposity and increased food and water intake, energy expenditure, and serum FGF21 levels compared to their relative controls. The RNAseq experiment was conducted to compare the effect of MR using these two protein sources on the hepatic transcriptome.

Implementation of dietary methionine restriction using casein after selective, oxidative deletion of methionine.

Project description:Dietary methionine restriction (MR) is normally implemented using diets formulated from elemental amino acids (AA) that reduce methionine content to ∼0.17%. However, translational implementation of MR with elemental AA-based diets is intractable due to poor palatability. To solve this problem and restrict methionine using intact proteins, casein was subjected to mild oxidation to selectively reduce methionine. Diets were then formulated using oxidized casein, adding back methionine to produce a final concentration of 0.17%. The biological efficacy of dietary MR using the oxidized casein (Ox Cas) diet was compared with the standard elemental MR diet in terms of the behavioral, metabolic, endocrine, and transcriptional responses to the four diets. The Ox Cas MR diet faithfully reproduced the expected physiological, biochemical, and transcriptional responses in liver and inguinal white adipose tissue. Collectively, these findings demonstrate that dietary MR can be effectively implemented using casein after selective oxidative reduction of methionine.

Effects of 20 months Dietary Methionine Restriction on Hepatic Gene Expression in Fischer 344 Rats

Project description:Dietary methionine restriction (MR) has been shown to increase lifespan and decrease adiposity in rodents. This study was designed to examine the transcriptional effects of MR in metabolically relevant tissues. This experiment contains data from the liver. We analyzed MR-induced changes in gene expression using pooled RNA from liver of rats fed either a control purified amino acid diet (DL-methionine content of 0.86%) (CON) or a methionine-restricted diet (DL-methionine content of 0.172%)(MR). Rats were fed Purina rodent diet 5001 until 32 days of age and were then randomly assigned to be fed CON diet or MR diet for 20 months.
