Project description:Random mutagenesis was applied to produce a new wheat mutant (RYNO3926) with superior characteristics regarding tolerance to water deficit treatment and rapid recovery from water stress conditions. Under water stress conditions mutant plants reached maturity faster and produced more seeds than its wild type wheat progenitor Further, whereas wild-type Tugela DN plants died within 7 days after induction of water stress, mutant plants survived by maintaining a higher relative moisture content (RMC), increased total chlorophyll, a higher photosynthesis rate and stomatal conductance suggesting that the mutant may possess a “stay green”’ phenotype. Analysis of the proteome of mutant plants revealed that they better regulate post-translational modification (SUMOylation) and have increased expression of ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) proteins. Mutant plants also expressed unique proteins associated with dehydration tolerance including abscisic stress-ripening protein, cold induced protein, cold-responsive protein, dehydrin, Group 3 late embryogenesis and also a lipoprotein (LAlv9) belonging to the family of lipocalins. Overall, our results suggest that our new mutant RYNO3936 has a potential for inclusion in future breeding programs to improve drought tolerance under dryland conditions.

Project description:We conducted microarray analysis to study comprehensive changes of gene expression profile under long-term low-temperature (LT) treatment and to identify other LT-responsive genes related with cold acclimation in seedling leaves and crown tissues (shoots containing apical meristems) of a synthetic hexaploid wheat line. The microarray analysis revealed marked up-regulation of a number of Cor/Lea genes and fructan biosynthesis-related genes under the long-term LT treatment. For validation of the microarray data, we selected four synthetic wheat lines, which contained the A and B genomes from a tetraploid wheat cultivar Langdon and the diverse D genomes originating from the different Ae. tauschii accessions, with distinct levels of freezing tolerance after cold acclimation. Quantitative RT-PCR analyses showed that the transcription accumulated levels of the Cor/Lea, CBF, and fructan biosynthesis-related genes were higher in more freezing-tolerant lines than those in the sensitive lines. The fructan biosynthesis pathway would be associated with cold acclimation to develop wheat freezing tolerance and related with diversity of the freezing tolerance level in addition to the CBF-mediated Cor/Lea expression pathway.

Two independent experiments were conducted in each exprement.

Project description:Microarrays were used to identify transcriptional responses in field-grown root material of wheat in order to dissect specific gene expression responses to limited macronutrient availability, particularly phosphate. This study fills the gap between the transcriptome studies on model plants and the lack of studies on soil-grown wheat aiming to identify candidate genes for enhancing nutrient uptake efficiency. The work at Rothamsted Research is supported via the 20:20 Wheat® Programme by the UK Biotechnology and Biological Sciences Research Council. The contribution was supported by BIONUT-ITN and the research leading to these results has received funding from the European Union Seventh Framework Programme (FP7/2007-2013) under grant agreement no. 264296. T. aestivum cv. Hereward root material was excavated in triplicates in May 2011 at booting stage from sections 0 and 1 plots representing continuous wheat plots of the “Broadbalk” field experiment at Rothamsted Research, UK (http://www.rothamsted.ac.uk/sample-archive/guide-classical-and-other-long-term-experiments-datasets-and-sample-archive). The distinct peculiarity of these plots, including a control plot with nutrient replete wheat plants, is the withdrawal of N, P, K, Mg and S fertilizers exposing the plants to multiple long-term nutrient deficiencies and representing 6 treatments ; 12 samples were analysed.
