Project description:We conducted an epigenome-wide association study (EWAS) to evaluate global DNA methylation in EOC. In a retrospective cohort of 80 primary ovarian cancers, we observed that global DNA hypermethylation was significantly associated with high-grade tumors, platinum-resistance and poor prognosis. We determined that 8,600 differentially methylated probes (DMPs) were enriched in the intergenic regions of 4,673 genes involved in small GTPases and were significantly correlated with the PI3K-AKT, MAPK, RAS, and WNT oncogenic pathways.

In a retrospective cohort of 30 HGSOC subtype primary ovarian cancers, we observed that global DNA hypermethylation was preferentially associated with recurrent HGSOC. A total of 2,969 DMPs corresponding to 1,471 genes were involved in olfactory transduction, and calcium and cAMP signaling.

Gene expression of papillary renal neoplasm with reverse polarity (PRNRP) cases + PRCC cases

Project description:Between 80% and 90% of renal carcinomas are either clear cell renal cell carcinoma or papillary RCC although there are at least 20 other distinct forms of RCC recognised by the World Health Organization (WHO). Emerging or provisional entities have been considered as variants of PRCC, including papillary renal neoplasm with reversed polarity (PRNRP). However, the molecular basis of PRNRP is unknown. We used microarray analysis to elucidate the gene profile of 10 PRNRP cases and an additional 7 PRCC cases. Cases were only considered for this study after fulfilment of strict inclusive histological and immunohistochemical criteria. We used Affymetrix Clariom D human arrays to elucidate the expression of these samples.
