Project description:Melatonin is a neurohormone that maintains the circadian rhythms of the body. By regulating the secretion of other hormones and neurotransmitters, it acts as a pleiotropic modulator that affects, for example, reproductive, immune, cardiovascular, sleep, and wake systems and mood. Thus, synthetic melatonin has become an essential component in the treatment of depressive disorders. Although we know the pathway of melatonin action in the brain, we lack comprehensive cross-sectional studies on the periphery of depressed patients. This study aimed to comprehensively analyze the differences between healthy control subjects (n = 84) and unipolar and bipolar depression patients (n = 94), including an analysis of the melatonin pathway at the level of the genes and serum biomarkers. An innovative approach is a pilot study based on gene expression profiling carried out on clinical and cell culture models using agomelatine and melatonin. We confirmed the melatonin biosynthesis pathway's molecular regulation dysfunctions, with a specific pattern for unipolar and bipolar depression, at the AANAT gene, its polymorphisms (rs8150 and rs3760138), and examined the serum biomarkers (serotonin, AANAT, ASMT, and melatonin). The biological pathway analysis uncovered pathways and genes that were uniquely altered after agomelatine treatment in a clinical model and melatonin treatment in a cell culture model. In both models, we confirmed the immunomodulatory effect of melatonin agents in depression.

Project description:Melatonin is a well-known agent that plays multiple roles in animals. Its possible function in plants is less clear. In the present study, we tested the effect of melatonin (N-acetyl-5-methoxytryptamine) on soybean growth and development. Both spraying of leaves and seed-coating with melatonin significantly promoted soybean growth as judged from leaf size and plant height. This enhancement was also observed in soybean production and their fatty acid content. Melatonin increased pod number, seed number and seed weight. However, the 100-seed weight was not influenced by melatonin application. Melatonin also improved soybean tolerance to salt and drought stresses. Transcriptome analysis revealed that melatonin up-regulated the expression of many genes and alleviated the inhibitory effects of salt stress on gene expressions. Further detailed analysis of the affected pathways documents that melatonin likely achieved its promotional roles in soybean through enhancement of genes involved in cell division, photosynthesis, carbohydrate metabolism, fatty acid biosynthesis and ascorbate metabolism. Our results demonstrate that melatonin has significant potential for improving of soybean growth and seed production. Further study should uncover more about the molecular mechanisms of melatoninM-bM-^@M-^Ys function in soybeans and other crops. Four different treatments were chosen, water, salt, 100M-BM-5M melatonin and salt plus 100M-BM-5M melatonin. The comparison of salt/melatonin-treated sample versus water-treated sample reveals salt or melatonin induced transcriptome changes. The comparison of melatonin plus salt treated sample versus salt-treated sample reveals melatonin induced changes when salt exists.

Project description:Melatonin, a natural phytohormone present in most plants, plays multiple roles in plant growth and stress responses. Although melatonin biosynthesis-related genes have been suggested to possess diverse biological functions, their roles and functional mechanisms in regulating rice grain yield remain largely unexplored. Here, we revealed that a rice caffeic acid O-methyltransferase (OsCOMT) gene is involved in melatonin biosynthesis through in vitro and in vivo evidences. Transgenic assays show OsCOMT significantly delays leaf senescence at the grain filling stage, and then improves photosynthesis efficiency. Further experimental and transcriptomic data suggest that OsCOMT inhibits the degradation of chlorophyll and chloroplast, which in turn delay leaf senescence. Histological analysis also reveals the role of OsCOMT in the development of vascular bundle system in rice. The levels of melatonin and cytokinin were significantly increased in the culm of OsCOMT-overexpression plant relative to those of the wild-type (WT). In the OsCOMT-overexpression line, the cytokinin-biosynthesizing genes were up-regulated and the cytokinin-degrading genes were down-regulated, thereby increasing the cytokinin levels compared with the WT. Thus, OsCOMT-mediated vascular patterning may result from the crosstalk between melatonin and cytokinin. More importantly, OsCOMT significantly increased grain number and yield production of rice in various background, including Nipponbare (NIP) and Suken118 (SK118). Our findings show novel insights into melatonin-mediated leaf senescence and vascular patterning, and provide a new strategy to enhance rice yield production.
