Project description:Adult male Sprague Dawley-derived rats were housed and fed as described previously (Dail et al., 2007). Three rats were treated i.p. with PB in saline at 80 mg/kg body weight/day (treated) for 5 days as described previously (Ma and Chambers; 1995). Following sacrifice on day 5, the livers were removed and stored at -80 C as previously described (Dail et al., 2007). Five 8 µm sections were taken from each of the three control and three experimental livers resulting in fifteen sections from each group using a Triangle Biomedical Supply Co Minotome Plus (Durham, NC) microtome. Differential detergent fractionation two dimensional liquid chromatography electrospray ionization tandem mass spectrometry (DDF 2D LC ESI MS2) and systems biology modeling were used to identify alterations in toxicologically relevant hepatic molecular functions and biological processes in the livers of rats

Project description:Canine mammary gland tumors can be used as predictive models for human breast cancer. There are several types of microRNAs common in human breast cancer and canine mammary gland tumors. The functions of microRNAs in canine mammary gland tumors are not well understood. In the present study, we compared the characterization of microRNA expression in two-dimensional and three-dimensional canine mammary gland tumor cell models. The expression of microRNA-210 in the three-dimensional-SNP cells was 10.19 times higher than that in the two-dimensional-SNP cells.

Project description:Purpose: The goals of this study are to analyze global transcriptional, epigenetic and topological changes in dendritic cells under LPS treatment condition and to investigate how CTCF regulates three-dimensional chromatin interaction in inflammatory response. Conclusions: Our study provides the detailed analysis of transcriptional, epigenetic and topological genome profiles in mouse dendritic cells under LPS treatment condition. Moreover, our data show how three-dimensional enhancer networks control gene expression during DC activation.
