Project description:Clinically important broadly reactive B cells evolve during multiple infections. The phenotype and function of B cells re-activated after secondary infection is different compared to B cells activated after a primary infection. Here we studied CD27highCD38high plasmablasts from patients with a primary or secondary dengue virus infection. Three transcriptionally and functionally distinct plasmablast clusters were identified. The largest cluster 0/1 was plasma cell-related and contained cells coding for virus serotype cross-reactive antibodies of the IgG1 isotype, consistent with memory B cell activation during an extrafollicular response. In contrast, cells in clusters 2 and 3 expressed low levels of antibody genes and high levels of genes associated with oxidative phosphorylation, EIF2 pathway and mitochondrial dysfunction. Clusters 2 and 3, which we name T cell-dependent plasmablasts, showed a transcriptional footprint of T cell help, in line with activation from either naÃƒÂ¯ve B cells or memory B cells. Our results contribute to the understanding of the parallel B cell activation events that occur in humans after natural primary and secondary infection.

Transcriptomic analysis of cell-of-origin CNS neuroblastoma, FOXR2 activated [Multiome]

Project description:The transcription factor FOXR2 is the universal driver of childhood central nervous system neuroblastoma, FOXR2 activated (NB-FOXR2). NB-FOXR2 tumors arise exclusively in the brain hemispheres, and despite morphological similarities to other pediatric brain tumors, they are a molecularly distinct entity based on DNA methylation profiling. The cell-of-origin is unknown. Here, we profiled a cohort of rare NB-FOXR2 tumors by bulk and single-cell transcriptomics. Through systematic comparative analyses, we delineate tumor transcriptional states and candidate cell-of-origin. More broadly, we demonstrate systematic molecular profiling of childhood cancers to orient oncogenic targeting for in vivo modeling, a critical resource for the study of rare tumors and development of therapeutics.

PD-1highCXCR5–CD4+ Peripheral Helper T (Tph) cells Promote CXCR3+ Plasmablasts in the early phase of COVID-19

Project description:T cell–B cell interaction is the key immune response to protect host from severe viral infection. We used COVID-19 as a model of acute viral infections and analyzed CD4+ T cell subsets associated with plasmablasts expansion and clinical outcome. Peripheral helper T cells (Tph, denoted as PD-1highCXCR5–CD4+ T cells) were significantly increased, as were plasmablasts. Tph cells exhibited “B-cell help” signatures and induced plasmablasts differentiation in vitro. Expanded plasmablasts showed increased CXCR3 expression, which is positively correlated with higher frequency of activated Tph cells and better clinical outcome. These results elucidate a critical role for Tph cells in regulating protective B cell response during acute viral infection.

CTCF binding in B cells and Plasmablasts

Project description:The mammalian CCCTC-binding factor (CTCF) regulates gene expression through the formation of higher order chromatin structures. Recent evidence has implicated a role for CTCF in regulating gene expression in the human MHCII locus. To investigate the role of CTCF in murine MHCII gene expression we mapped CTCF binding sites in B cells (MHCII+ cells) and Plasmablasts which are differentiated B cells that have silenced MHCII gene expression. These observations lead to the identification of differential CTCF binding during differentiation in these cell types and suggest mechanims of MHCII gene regulation.
