Project description:Lettuce (Lactuca sativa L.) is a highly perishable horticultural crop with a relatively short shelf life due to leaf senescence that limits its commercial value and contributes to food waste. Postharvest senescence varies with influences of both environmental and genetic factors. Preharvest genetic factors can be indicative of postharvest quality. Discovery of additional preharvest markers to assess lettuce shelf life is an important step towards increasing the efficiency of lettuce breeding efforts for improved shelf life. We selected and evaluated three romaine lettuces with variable shelf lives with the aim of identifying preharvest markers of lettuce postharvest shelf life. We evaluated leaf morphological characteristics for each of the three cultivars. To assess molecular indicators of shelf life, we used an RNA sequencing approach to construct transcriptomic profiles of two of the cultivars, a short shelf life (SSL) breeding line 60184 and a long shelf life (LSL) cultivar ‘Okeechobee’ at maturity. We identified 552 upregulated and 315 downregulated differentially expressed (DE) genes between the genotypes. We found that 27 % of the DE lettuce genes had an Arabidopsis thaliana ortholog characterized as senescence-associated, indicating that variable expression of senescence-associated genes (SAGs) could serve as a tool for preharvest markers of postharvest shelf life. Notably, we identified several SAGs and functional groupings with highly differential expression between the cultivars. This includes several jasmonate ZIM-domain (JAZ), jasmonic acid (JA) signaling genes, chlorophyll a-b binding (CAB) chloroplast-associated genes, and cell wall modification genes including pectate lyases (PL) and expansins (EXP). This study presented an innovative approach for identifying molecular markers for preharvest factors linked to postharvest traits for prolonged shelf. Project description:Enterobacter sp. SA187 is a plant growth-promoting bacterium (PGPB) that promotes growth of the crop plant alfalfa under saline irrigation and desert farming conditions. SA187 also enhances salt tolerance of the model plant Arabidopsis thaliana under in vitro conditions. In the present study, we used a transcriptomic approach to elucidate the mechanisms underlying plant growth promotion by SA187 under salt stress. Compared to free-living SA187, a massive metabolic reprogramming of SA187 occurs upon association with Arabidopsis. This effect was largely independent of the plant growth condition (non-salt or salt stress). Our data revealed pronounced changes in gene expression of proteins involved in cell signaling, chemotaxis, flagella biosynthesis, quorum sensing and biofilm formation. Also, upon plant interaction, a complete reprograming of nutrients acquisition and the central carbon metabolism of SA187 was observed. Moreover, in accordance with the previously identified role of bacterially produced 2-keto-4-methylthiobutyric acid (KMBA) in mediating salt stress tolerance, the sulfur metabolism of SA187 was strongly induced. Overall, our results give a deep insight into the metabolic and signaling pathways involved in the transition from free-living to a plant-associated PGPB life style of SA187.

Project description:Passiflora edulis, commonly known as passion fruit, is a crop with a fragrant aroma and refreshingly tropical flavor that is a valuable source of antioxidants. It offers a unique opportunity for growers because of its adaptability to tropical and subtropical climates. Passion fruit can be sold in the fresh market or used in value-added products, but its postharvest shelf life has not been well-researched, nor have superior cultivars been well-developed. Understanding the proteins expressed at the tissue level during the postharvest stage can help improve fruit quality and extend shelf life. In this study, we carried out comparative proteomics analysis on four passion fruit tissues, the epicarp, mesocarp, endocarp, and pulp, using multiplexed isobaric tandem mass tag (TMT) labeling quantitation. A total of 3,352 proteins were identified, including 295 differentially expressed proteins (DEPs). Among the DEPs, there were proteins expressed with functions in oxygen scavenging, lipid peroxidation, response to heat stress, and pathogen resistance. This research provides insight into tissue-specific pathways that can be further studied within fruit physiology and postharvest shelf life to aid in implementing effective plant breeding programs. Knowing the tissue-specific function of fruit is essential for improving fruit quality, developing new varieties, identifying health benefits, and optimizing processing techniques.
