Comparison of Apis mellifera workers and queens hind leg development at prepupal stages

Project description:In Apis mellifera, the female eggs can develop into workers or queen depending on the diet offered during early development. The outputs of the developed honeybee females are two morphs with particular morphological traits and related physiology. Among the specialized structures in workers the hind tibia forms the corbicula or the pollen basket, a smooth region surrounded by a row of a long scopal hairs, used for carrying pollen and other materials to the nest. This morphological trait and the respective behaviour are absent in queens. Herein we show details of the initial steps of hind legs morphogenesis in honeybee castes. Using results from the hybridization of whole genome-based oligonucleotide arrays with RNA samples from hind leg imaginal discs of pre-pupa honeybees of both castes we present a list of differentially expressed genes.

Expression profile of third instar larvae tissues from wild type and bsAS mutant flies

Project description:Our data on third instar larvae tissues of Drosophila melanogaster has allowed us to identify bsAS, a lncRNA antisense to the blistered/DSRF gene, orthologous to the SRF transcription factor in humans, mainly expressed in fly wings. bsAS controls the expression of different isoforms of the blistered gene in Drosophila tissues. CRISPR-CAS deletion of bsAS Transcription Start Site thoroughly abolishes bsAS transcription, inducing a change in the bs isoform usage and the impairment of wing development. Our data consists on stranded RNA-Seq from eye-antenna, leg and wing imaginal discs from third instar larvae. To analyse bsAS mutation, we also performed stranded RNA-Seq of wild type and bsAS-/- eyes and wings from third instar larvae and late pupae.

A common set of DNA regulatory elements shapes Drosophila appendages

Project description:We mapped open chromatin by FAIRE-seq and measured gene expression by RNA-seq in 3 types of Drosophila samples: staged whole embryos, imaginal discs, pharate appendages. We first demonstrate that regions of open chromatin precisely define regions of enhancer activity in developing embryos. In contrast to the dynamic changes in open chromatin observed between different stages of embryogenesis, we found that the open chromatin profiles in wing, leg, and haltere imaginal discs are nearly identical. This was also true again later in development, where the adult appendages also share nearly identical open chromatin profiles. Therefore, at a given developmental time point, different appendages are specified using a shared set of DNA regulatory elements. However, from one time point to the next, the set of accessible regulatory elements changes. Open chromatin profiles in appendage imaginal discs are almost entirely different than those of the adult appendages. We propose that master regulator transcription factors create morphologically distinct structures by differentially influencing the function of the same set of DNA regulatory modules. Open chromatin profiling during Drosophila development: 3 stages of embryogenesis (2-replicates each); wing, leg, and haltere 3rd instar imaginal discs (3-replicates each); 3rd larval central nervous system (2-replicates); eye-antennal imaginal discs (2-replicates); wing, leg, and haltere pharate appendages (2-replicates each); Genomic DNA Inputs.
