Project description:Myxococcus xanthus and Escherichia coli represent a well-studied microbial predatorprey pair frequently examined in laboratory settings. While significant progress has been made in comprehending the mechanisms governing M. xanthus predation, various aspects of the response and defensive mechanisms of E. coli as prey remain elusive. In this study, the E. coli large-scale chromosome deletion library was screened, and a mutant designated as ME5012 was identified to possess remarkable resistance to predation by M. xanthus. Within the deleted region of ME5012 encompassing seven genes, the significance of dusB and fis genes in driving the observed resistant phenotype became apparent. Specifically, the deletion of fis resulted in a notable reduction in flagellum production in E. coli, contributing to a certain level of resistance against predation by M. xanthus. Meanwhile, the removal of dusB in E. coli led to diminished inducibility of myxovirescin A production by M. xanthus, accompanied by a slight increase in resistance to myxovirescin A. These findings shed light on the molecular mechanisms underlying the complex interaction between M. xanthus and E. coli in a predatory context.

Project description:In response to starvation Myxococcus xanthus initiates a developmental program that culminates in the formation of fruiting bodies inside which the rod-shaped cells differentiate to spores. Fruiting body formation depends on intercellular communication and two intercellular signals are known, the A-signal and the C-signal. Five genes have been identified which are required for A-signal synthesis. To begin to understand the function of the genes required for A-signal synthesis, we have analysed gene expression in the asgA and the asgB mutant. Project description:Phosphate regulation is complex in the developmental prokaryote Myxococcus xanthus, and requires at least three previously characterized two-component systems (TCS), designated PhoR1-PhoP1, PhoR2-PhoP2, and PhoR3-PhoP3. We report here the identification and characterization of a member of a fourth TCS, designated PhoP4, which shows high sequence similarity to the other three M. xanthus PhoP response regulators. PhoP4 is an orphan response regulator that was identified by yeast 2-hybrid screen as a possible interacting partner of PhoR2. phoP4 insertion mutation and in-frame deletion strains were constructed and assessed for developmental and phosphatase phenotypes, and then compared with the phenotypes previously reported for a _phoR2-phoP2 strain. The data indicate that the phoP4 mutations caused spore viability to be decreased by nearly two orders of magnitude, and reduced all three development-specific phosphatase activities by 80-90% under phosphate-limiting conditions. These phenotypes are stronger than those observed for mutations in the PhoR2-PhoP2 system, thereby suggesting a high degree of complexity vis-à-vis the interactions amongst the Pho signal transduction pathways in M. xanthus.
