Project description:It is generally assumed that parental DNA imprinting is complete before the onset of meiosis in human spermatogenesis. Conversely, we here demonstrate dynamic DNA methylation in multiple imprinted regions during meiosis and subsequent maturation of spermatids. This suggests a role for these genes in spermatogenesis and has implications for the effectiveness and safety of assisted conception using testicular sperm.

Project description:Mammalian parental imprinting represents an exquisite form of epigenetic control regulating the parent-specific monoallelic expression of genes in clusters. While imprinting perturbations are widely associated with developmental abnormalities, the intricate regional interplay between imprinted genes makes interpreting the contribution of gene dosage effects to phenotypes a challenging task. Using mouse models with distinct deletions in an intergenic region controlling imprinting across the Dlk1-Dio3 domain, we link changes in genetic and epigenetic states to allelic-expression and phenotypic outcome in vivo. This determined how hierarchical interactions between regulatory elements orchestrate robust parent-specific expression, with implications for non-imprinted gene regulation. Strikingly, flipping imprinting on the parental chromosomes by crossing genotypes of complete and partial intergenic element deletions, rescues the lethality of each deletion on its own. Our work indicates that parental origin of an epigenetic state is irrelevant as long as appropriate balanced gene expression is established and maintained at imprinted loci.

Project description:A reporter transgene displayed parental imprinting in mouse embryos when positioned into the Itga6 gene. The strong lacZ pattern of expression scored in embryos inheriting the transgene from a male was not present when transmitted from a female. The transgene exhibited maternal allele-specific DNA hyper-methylation acquired in the germ-line and histone modifications corresponded to profiles described at known imprinted loci. Chromosome conformation analyzes revealed distinct, parent-of-origin interaction domains, with a more compact structure characterizing the maternally inherited repressed allele. The analysis of such transgene insertions with a selective potential to induce imprinting may help understanding the mechanisms identifying particular loci as targets for allele-specific repression.

Parental allele specific single-cell transcriptome dynamics reveal incomplete epigenetic reprogramming in human female germ cells

Project description:In contrast to mouse, human female germ cells develop asynchronously. Germ cells transition to meiosis, erase genomic imprints, and reactivate the X chromosome. It is unknown if these events all appear asynchronously, and how they relate to each other. Here we combine exome sequencing of human fetal and maternal tissues with single-cell RNA-sequencing of five donors. We reconstruct full parental haplotypes and quantify changes in parental allele specific expression, genome-wide. First we distinguish PGC, pre-meiotic, and meiotic transcriptional stages. Next we demonstrate that germ cells from various stages monoallelically express imprinted genes from e.g. the SNURF-SNRPN cluster and confirm it by methylation patterns. Finally we show that ±30% of the PGCs, are still reactivating their inactive X chromosome and that this is related to transcriptional stage, and not to embryonic age. Altogether, we reveal the complexity and cell-to-cell heterogeneity of transcriptional and epigenetic remodelling in female human germ cells.
