Project description:The most commonly used inbred mouse strain, C57BL/6J, lacks a functional nicotinamide nucleotide transhydrogenase (Nnt) and thus, is protected from pressure-overload-induced oxidative stress and heart failure. We screened for differential gene expression in left ventricular myocardium after sham/transverse aortic constriction (TAC) surgery in 10-12 weeks old mice from BL/6N (functional Nnt) and BL/6J (non-functional Nnt, missense of exons 7-11) strains.
Project description:A number of mediators that have been found to be involved in the pathogenesis of cardiac hypertrophy affecting gene transcription, protein synthesis, metabolism,autophagy, oxidative stress and inflammation. These mediators including nuclear transcription factor, microRNAs and long noncoding RNAs. However, the gene expression profiles signaling pathways in the pressure overload-induced cardiac hypertrophy remain unknown. All mice were randomly undertaken sham surgery or TAC for 2 weeks. Hearts were harvested 2 weeks following TAC and performed to further analysis.
Project description:4 Adult male Sprague-Dawley rats (275-350 g) were anesthetized and subjected to hepatectomy sham surgery (abdominal cavity was opened, liver was handled, but no tissue resection was made). 1 hour after the surgery rats were killed and liver samples were harvested. This study was conducted to analyzes the effects of surgical stress on gene expression levels in rat liver. It provides additional data to 1-6 h partial hepatectomy study (Series GSE7415). Project description:whole gene expression analysis for ZBTB17 cKO mice after transverse aortic constriction (TAC) We used microarrays to detail the global programme of gene expression underlying hypertrophy and heart failure process upon pressure overload 16 male mice, 4 mice per group, were selected at 8-10 weeks old for 4 weeks of TAC and SHAM and collected samples for RNA extraction and hybridization on Affymetrix microarrays. Project description:PIEZO1 is a mechanically-activated ion channel that contributes to flow sensing in vascular endothelium. Moreover, deletion of endothelial PIEZO1 was recently found to suppress activation of Notch1 target genes in hepatic microvascular endothelium. Here, because of the liver’s dominant role in lipid regulation, we set out to test the novel hypothesis that endothelial PIEZO1 regulates hepatic lipid homeostasis. We performed bulk RNA sequencing on PIEZO1-deleted mice exposed to chow and high fat diets. Our transcriptomics analysis reveal unexpected relevance to lipid and glucose homeostasis.
