Project description:We used Next-generation sequencing (NGS) to reveal MbovP475-binding sites in BoMac cells. We extracted the genome of BoMac infected with mycoplasma bovis by sonication, then the sheared chromatin in the supernatant was incubated with purified rabbit polyclonal IgG against MbovP475 and protein A/G magnetic beads. The DNA was eluted from the beads and subject to making sequencing library. We totally got 2648 potential MbovP475-binding sites by ChIP-seq. Among them, CRYAB and MCF2L2 gene were further verifed to be down-reguated by MbovP475 through binding to the promoter region of two genes.
Project description:We report the high-throughput profiling of ISL1-binding sites in mammalian cells. By obtaining over four billion bases of sequence from chromatin immunoprecipitated DNA, we generated genome-wide ISL1-binding maps of Huh7 cells. This study provides a prediction of regulated genes by the ISL1.
