Project description:The liver plays a central role in vertebrate glucose homeostasis, and is also one of the most sexually dimorphic organs in terms of gene expression. While the extent of hepatic sexual dimorphism has been well described in mammals, little is known regarding this phenomenon in non-mammalian species, particularly fish. In this study, we examined hepatic gene expression and physiological phenotypes (growth, proximate body composition, retention efficiencies) to determine whether male and female zebrafish respond differently to diets comprised of 0, 15, 25, or 35 % carbohydrate. Using both Affymetrix microarrays and qRTPCR, we observed substantial sexual dimorphism in the hepatic transcriptome, and the response of some genes to dietary carbohydrate manipulation also varied by sex. Males upregulated genes associated with oxidative metabolism, carbohydrate metabolism, energy production, and amelioration of oxidative stress, while females had higher expression levels of genes associated with translation. Males also expressed elevated levels of hnf4α, a gene thought to be involved in regulating hepatic sexual dimorphism in the rodent. Dietary carbohydrate affected hepatic gene expression, growth performance, retention efficiencies of protein and energy, and percentage of moisture, lipid, and ash. Significant diet effects reflected differences between the 0% carbohydrate diet and the other diets, consistent with previous work on other cyprinids showing a high tolerance for dietary carbohydrate. Project description:Mandarin fish Siniperca chuatsi (Basilewsky) (Percichthyidae), as a demersal piscivore, has very specialized feeding habits, for as soon as they start feeding the fry of this fish feed solely on fry of other fish species. In rearing conditions, mandarin fish has been found to accept live prey fish only, and refuse dead prey fish or artificial diets, very little is currently known about the molecular mechanisms of multiple genes which cover different pathways influencing the specialized food habit, such as live prey. We performed transcriptome comparisons between dead prey fish feeders and nonfeeders in mandarin fish. The determination mechanisms of specialized food habit (live prey fish) in mandarin fish could provide some instructions for research of food habit in animals, including mammals.

Project description:The inclusion of intact phospholipids in the diet is essential during larval development and can improve culture performance of many fish species. The effects of supplementation of dietary phospholipid from marine (krill) or plant (soy lecithin) sources were investigated in Atlantic salmon, Salmo salar. First feeding fry were fed diets containing either krill oil supplying phospholipid at 2.6% of diet (named K2.6) or soybean lecithin supplying phospholipid at 2.6 % (S2.6), 3.6 % (S3.6) of diet. A control diet (B) without supplemented phospholipid was also supplied. Fish were sampled at ~ 2.5 g (~1990 ˚ day post fertilization, dpf) and ~10 g (2850 ˚dpf). By comparison of the intestinal transcriptome in specifically chosen contrasts, it was determined that by 2850˚dpf fish possessed a profile that resembled that of mature and differentiated intestinal cell types with a number of changes specific to glycerophospholipid metabolism. It was shown that intact phospholipids and particularly phosphatidylcholine are essential during larval development and that this requirement is associated with the inability of enterocytes in young fry to endogenously synthesize sufficient phospholipid for the efficient export of dietary lipid. In the immature phase (~1990 ˚dpf), the dietary phospholipid content as well as its class composition impacted on several biochemical and morphological parameters including growth, but these differences were not associated with differences in intestinal transcriptomes. The results of this study have made an important contribution to our understanding of the mechanisms associated with lipid transport and phospholipid biosynthesis in early life stages of fish.

Project description:Food is a fundamental requirement for organisms to develop, survive and reproduce. However, access to an optimum diet is not always guaranteed, resulting in a mismatch between the diet and the organism's needs. To comprehend the consequences of this mismatch, it becomes crucial to explore how different compositions of an organism's diet impact its overall performance. In this context, we tested the effect of isocaloric diets with different protein to carbohydrate ratios on eight key life history traits in a large (n = ~2,100) outbred Drosophila melanogaster population. We report accelerated pre-adult development and increased reproductive output, without affecting pre-adult viability and body size, in flies fed on low carbohydrate diet. We also observe that high carbohydrate content in diet shortens fly lifespan. In support, we found that increasing carbohydrate content in food led to faster aging by impairing locomotor activity and gut integrity. Additionally, we noted that the composition of the diet had an impact on gene expression patterns with high carbohydrate diets upregulating genes and high protein diets downregulating them. These results collectively provide a comprehensive understanding of how the balance of macronutrients in an organism's diet intricately shapes its traits.
