Project description:Transcriptional profiling of three mexican maize landraces under 10, and 17 days stress and recovery irrigation A dye balanced modified loop design was implemented. Two biological replicates (pooling five representative plants) representing each sampling point for each genotype were obtained for purified RNA from 120 randomly chosen seedlings. This experiment involved a total of forty-eight (24 sets) of microarray hybridizations, including direct and dye swap comparisons between treatments as well as across the three landraces. This design allowed us to determine differences in gene expression between the three different landraces under drought stress (10 and 17 days) and at recovery irrigation compared to irrigated controls.

Project description:<p>The intensity and frequency of precipitation events are predicted to change over the coming decades. For many areas, longer periods without rainfall are expected. We investigated the importance of irrigation frequency under water deficit conditions for growth, physiology and chemistry of wheat (<em>Triticum aestivum</em>). Drought-stressed plants received 40% of the water provided for control plants and were either watered every other day (continuous drought, cd) or every eight days (pulsed drought, pd). Maximum quantum yield of photosystem II (Fv/Fm), aboveground biomass, applied water use efficiency (WUEapl) and the flag leaf metabolome were assessed twice during development. Fv/Fm was not affected by irrigation. Drought-exposed plants produced less biomass, but had higher WUEapl than control plants. More metabolic features responded to the pd compared to the cd treatment and more features were increased than decreased in pool size in flag leaves. Salicylic acid glucoside was generally decreased under drought. In pd plants, two benzoxazinoid glucosides were enhanced at the first time point and concentrations of several flavonoid glycosides were modulated. This study extends our knowledge about drought effects on wheat; it highlights that the frequency of watering determines how plant growth, physiology and metabolism are affected by drought.</p>

Project description:The knowledge of proper fertigation across various irrigation levels is necessary for maximizing peanut yield and irrigation use efficiency in arid areas, and it also can effectively alleviate the risk of nutrient deficiency induced by water stress. This study evaluated the effectiveness of cobalt combined with two zinc application methods on peanut nutrient uptake, yield, and irrigation water use efficiency across varying irrigation levels. A split-split plot experiment was carried out in 2021 and 2022. Three peanut gross water requirement (GWR) levels (100%, 80%, and 60%) were designated for main plots. Subplots featured plants treated with either 0 or 7.5 mg L-1 of cobalt. The sub-sub plots assessed chelated zinc effects at rates of 0 and 2 g L-1 via foliar and soil applications. In comparison to the control (100% GWR), nutrient uptake decreased, with sodium being the exception, and there was an increase in soil pH at 60% GWR. The results showed also significant reductions in yield and water use by approximately 60.3% and 38.1%, respectively. At this irrigation level, applying zinc via soil, either alone or combined with cobalt, led to significant yield increases of 89.7% and 191.3% relative to the control. Also, it's crucial to note that cobalt application negatively affected iron and copper at 60% GWR, but this impact was lessened with soil-applied zinc. Hence, under a similar circumstance, treating stressed peanut plants with additional foliar applications of iron + copper and applying zinc via soil, could enhance nutrient uptake and improve yield. On the other hand, at 80% GWR, a combination of foliar-applied zinc and cobalt, had a tremendous impact on the absorption of (nitrogen, phosphorus, magnesium, and zinc), resulting in enhanced agronomic traits and decreased water losses. Additionally, at this irrigation level, foliar zinc application alone yielded a 32.4% increase compared to the 80% GWR control. When combined with cobalt, there was a 70.0% surge in water use. Based on this knowledge, the study suggests using 80% GWR and treating peanut plants with a combination of foliar-applied zinc and cobalt. This strategy aids plants in countering the adverse effects of water stress, ultimately leading to enhanced yield and irrigation water use efficiency.

Project description:Changes in gene expression during berry development during a grape growing season were analysed. Project description:A mid-thoracic aorta coarcted rat was created to generate a stable pressure difference above versus below the coarctation ligature. This study determined that the PVAT around the thoracic aorta exposed to a higher pressure has a significantly reduced ability to assist stress relaxation versus that below the ligature and appears to retain the ability to be anticontractile. At the same time, the PVAT around the thoracic aorta exposed to higher pressure had a reduced adipogenic potential versus that below the ligature. Transcriptomics analyses indicated that PVAT below the coarctation showed the greatest number of DEGs with an increased profile of the synaptic neurotransmitter gene network.

Project description:Deficit irrigation (DI) was acknowledged as an effective technique to improve water use efficiency (WUE) without significant yield reduction. In this study, a 3-year field experiment was conducted in Northeast China during 2017-2019 to investigate the combined effects of 3-week DI from 3-leaf stage and N fertilization on maize seedling growth and determine the resulting impacts on silking growth and yield formation, N use efficiency (NUE) and WUE. Results showed that seedling-stage DI decreased leaf area and photosynthesis, thus significantly limited shoot and root dry biomass for maize seedling, compared to well-watered (WW) plants. In 2017 and 2019, seedling-stage DI positively improved seedling growth with higher root: shoot ratio and enhanced drought tolerance, under higher initial soil water contents (SWC) with sufficient precipitation before DI. The DI-primed plants showed similar or better performances on reproductive growth, grain yield, WUE and NUE compared to WW plants, even experiencing heavy rainfall or drought stresses around the silking stage. However, the contrasting results were observed in 2018 with negative DI effects on seedling and silking growth and final yield, probably due to less rainfall and lower SWC before DI. In all 3 years, N fertilization had significant compensatory effects on limited seedling growth under DI, and its effect was much less in 2018 than other years due to adverse early climate. The principal component and correlation analysis revealed maize silking growth, grain yield, NUE and WUE were strongly related to the seedling growth as affected by water and N managements under various climatic conditions. In conclusion, a short-term and moderate DI regime-adopted at the seedling stage under higher initial SWC and coupled with an appropriate N fertilization-is beneficial to control redundant vegetative growth while optimizing root development, therefore effectively improving drought tolerance for maize plants and achieving higher grain yield, WUE and NUE.
