Project description:Congenital heart disease is a congenital defect with a very high morbidity and mortality rate, of which Cyanotic Congenital Heart Disease (CCHD) is the most severe type.Due to the special anatomical changes of CCHD, the blood of the right ventricle cannot enter the pulmonary capillaries for full oxygenation, directly flows into the left ventricle and further flows into the systemic circulation, resulting in long-term hypoxia.Therefore, the study of the injury mechanism of cardiomyocytes under chronic hypoxia may provide a new treatment strategy for patients with cyanotic coronary heart disease.Mouse cardiomyocytes(CMCs) were treated with hypoxia for different periods of time (oh, 24h, 48h and 72h, respectively), followed by label free proteomic analysis.

Large-scale non-targeted serum metabolomics in the Swedish Twin Registry

Project description:The TwinGene project, conducted between 2004 and 2008, is a population-based Swedish study of twins born between 1911 and 1958. The study participants have previously participated in a telephone interview called Screening Across the Lifespan Twin Study, conducted between 1998 and 2002. To be included in TwinGene, both twins within a pair had to be alive. In total, 12591 individuals participated by donating blood to the study, and by answering questionnaires about life style and health. For the purpose of metabolomic profiling, we designed a case-cohort of incident coronary heart disease, diabetes, dementia and ischemic stroke events and a matched sub-cohort (controls) stratified on age and sex. In the final analysis we included serum samples from 2,139 individuals.

Project description:Background:Femaleinfantswith congenital heart disease (CHD)facesignificantlyhigherpostoperativemortality ratesafter adjusting for cardiac complexity.Sex differences in metabolic adaptation to cardiac stressors maybe an earlycontributor tocardiac dysfunction.In adult diseases, hypoxic/ischemic cardiomyocytesundergo acardioprotectivemetabolicshift fromoxidative phosphorylation to glycolysiswhichappears to be regulated in a sexually dimorphic manner.We hypothesizesex differences incardiacmetabolismare presentincyanotic CHDand detectable asearly as theinfantperiod. Methods:RNA sequencingwas performedon blood samples(cyanotic CHD cases,n=11;controls,n=11)andanalyzedusinggene set enrichment analysis (GSEA). Global plasma metaboliteprofiling(UPLC-MS/MS)was performedusinga larger representative cohort(cyanotic CHD,n = 27;non-cyanotic CHD,n = 11;unaffectedcontrols,n = 12). Results:Hallmark gene sets in glycolysis, fatty acid metabolism, and oxidative phosphorylationwere significantly enrichedincyanotic CHDfemales compared to malecounterparts, which was consistent withmetabolomicdifferences between sexes.Minimalsex differencesinmetabolicpathwayswereobservedinnormoxicpatients(bothcontrols and non-cyanotic CHDcases). Conclusion:These observationssuggestunderlying differences inmetabolicadaptationtochronic hypoxiabetween males and females withcyanotic CHD.

Project description:Tetralogy of Fallot (TOF) is the most common cyanotic congenital heart defect with a world-wide prevalence of 3 to 4 cases per 10,000 life births. TOF is a congenital heart disease with four major cardiac defects, i.e., ventricular septal defect, overriding aortic root, infundibular stenosis of the pulmonary artery, and right ventricular hypertrophy. Treatment relies on correction surgery in early infancy. This study performed whole genome microarray gene expression profiling of cardiac specimens of the right ventricular outflow tract (RVOT), which were recovered during correction surgery of TOF from 11 pediatric patients diagnosed with TOF cardiac defects.

Congenital scolisis may be associated with both of rare mutation and allele-specific methylation

Project description:Congenital scoliosis (CS) is a congenital disease that results in anomalous vertebral development. Research indicates that both genetic and environmental factors during pregnancy contribute to the risk of CS development, however, the exact mechanisms underlying CS pathogenesis remain unclear. To address this issue, we have generated both genetic (by exon capture sequencing) and epigenetic (by MeDIP) maps from a CS disease-discordant monozygotic twin pair. We compared the differences in the presence of common and rare SNPs between the twins as well as investigated differences in methylation patterns. Our data indicated that rare mutations are more likely to underlie CS development than common mutations, and that these are also more likely to play a role than do genomic methylation patterns. We did, however, identify a difference in the allele-specific-methylation between the twins in the SVIL gene which has functions in categories relevant to this disease, which raises the possibility of a novel mechanism promoting CS pathogenesis.
