Project description:We identified and functionally explored D. silvarum miRNAs involved in cold response to gain further understanding of the molecular regulatory mechanisms underlying the cold stress in ticks. The microRNA libraries of D. silvarum were established via high-throughput sequencing after exposure to different cold treatments. A total of 147 miRNAs, including 44 known miRNAs and 103 new miRNAs, were identified.

Project description:Ticks are vectors of different pathogens causing human and animal diseases. Particularly, Rickettsia slovaca is zoonotic infectious bacterium transmitted by Dermacentor ticks, agent of tick-borne lymphadenopathy (TIBOLA), common across Europe. Current studies point to extreme complexity of bacterial induced effects in tick host. Systems biology tools, including proteomics, greatly contribute to understanding of molecular details of pathogen-tick-host interactions. Herein we compared laboratory-infected ticks with uninfected control after four weeks of incubation. Propagation of R. slovaca was confirmed by quantitative PCR. Using DNA was confirmed infection with R. slovaca. By proteomic approach, we discovered 33 differentially abundant gel spots, 23 of them accumulated upon artificial infection with R. slovaca. Modest 6.9% of tick proteome was affected. The protein localizations showing that eight proteins spots might be secreted, three cytoplasmic, two mitochondrial, six likely having multiple localizations, one cell membrane and one nucleus. We identified following proteins defensin, serpins, glycine-rich protein, heat shock protein involved in artificially infected tick vector, Dermacentor reticulatus. Discovered differentially abundant proteins should be further evaluated as targets to block the transmission of bacterial pathogen.

Project description:The Ixodidea tick Dermacentor marginatus is a vector of many pathogens wide spread in Eurasia. Study of gene targets of the tick species provides insight to find novel tick protective antigen for drug development and vaccine targets. To obtain a broader picture of gene sequences and changes in expression level, we aimed to characterize the whole body transcriptome in D. marginatus adult female after engorgement and long-term starvation using RNA-seq. We have assembled and analyzed transcriptome of D. marginatus females 5 days after ecdysis, 24 h after a blood meal, and 6 months under controlled experimental conditions. Sequencing produced 30251 unigenes, of which 32% were annotated using Trinity. Gene expression was compared among groups differed by status as newly molted, starved and engorged female adult ticks. Nearly 1/3 of the unigenes in each group were differentially expressed compared to the other two group, and we found that the most numerous were proteins involved in catalytic and binding activities and apoptosis. Selected up-regulated differentially expressed genes in each group associated to protein, lipids, carbohydrate and chitin metabolism. Blood feeding and long-term starvation also caused genes differentially expressed in the defense response and antioxidant response. Finding the sequence information and expression pattern would be helpful in understanding molecular physiology of D. marginatus, and provides information for anti-tick vaccine and drug development.

Project description:BACKGROUND: Ticks are a kind of external parasitic arthropods that can transmit a variety of pathogens by sucking blood. Low temperature tolerance is an important guarantee for ticks to survive the cold winter. Exploring the protein regulation mechanism of low temperature tolerance of Haemaphysalis longicornis could help to explain how ticks survive in cold winter. At the same time, it has certain theoretical guiding significance for how to use winter to better control tick population. RESULTS: In this study, the quantitative proteomics of several tissues of Haemaphysalis longicornis exposed to low temperature was studied by DIA technology. 3699, 3422, and 1958 proteins were identified in salivary gland, midgut, and ovary, respectively. The proteins involved in energy metabolism, cell signal transduction, protein synthesis and repair, and cytoskeleton synthesis changed under low temperature stress. After a variety of bioinformatics analysis, it was found that arylamine N-acetyltransferase, inositol polyphosphate multikinase, and dual specificity phosphorase were of great significance in low temperature tolerance. Then we performed RNA interference on the mRNA of these three proteins, and the results showed that the ability of female ticks to tolerate low temperature decreased significantly. CONCLUSION: The comprehensive analysis of the protein regulation of multiple tissues of female ticks exposed to low temperature showed that maintaining cell homeostasis, maintaining cell viability, and enhancing cell tolerance were the most important means for ticks to maintain vital signs under low temperature. The expression of proteins involved in and regulating the above cell activities was the key to the survival of ticks under low temperature.

Project description:Recently, intensive global climate change has become a major factor impacting plant survival during the winter. Freezing cold temperatures during the winter and abnormal temperature fluctuations during the winter and early spring are the most harmful ambient factors threatening tea plant winter survival and currently cause marked economic losses in tea production. In this study, by simulating natural climate change, we established cold acclimation (CA) and rapid cold stress (after CA) conditions to comprehensively investigate the transcriptome changes involved in CA and rapid cold stress. Electrolyte leakage (EL) rate and expression profile clustering analyses confirmed that the experimental design was valid. Comparative transcription analysis identified many differentially expressed genes (DEGs) involved in both processes. Time course and pathway enrichment analyses further revealed the physiological changes that occur during the initial period of CA and the cell wall changes that occur throughout the entire CA process; these changes play crucial roles in increasing freezing tolerance during this process. Compared with CA, different cold response mechanisms were rapidly activated under cold stress; however, the subsequent accumulation of reactive oxygen species, which affect multiple aspects, caused by freezing cold could be the harshest factor impairing tea leaves. Moreover, we investigated 60 DEGs shared by both processes and highlighted the importance of KCSs, HXXXD-type acyl-transferase family proteins, NAC080, SWEETs and ENOs in the responses to various cold conditions. These results greatly improve our knowledge of cold response mechanisms in tea plants and provide meaningful information for functional studies investigating cold tolerance-related genes.
