Differential regulation of flower transpiration during abiotic stress in annual plants.

Project description:Heat waves occurring during droughts can have a devastating impact on yield, especially if they happen during the flowering and seed set stages of the crop cycle. Global warming and climate change are driving an alarming increase in the frequency and intensity of combined drought and heat stress episodes, critically threatening global food security. Because high temperature is detrimental to reproductive processes, essential for plant yield, we measured the inner temperature, transpiration, sepal stomatal aperture, hormone concentrations and transcriptomic response of closed soybean flowers developing on plants subjected to a combination of drought and heat stress. Here, we report that, during a combination of drought and heat stress, soybean plants prioritize transpiration through flowers over transpiration through leaves by opening their flower stomata, while keeping their leaf stomata closed. This acclimation strategy, termed 'differential transpiration', lowers flower inner temperature by about 2-3°C, protecting reproductive processes at the expense of vegetative tissues. Manipulating stomatal regulation, stomatal size and/or stomatal density of flowers could serve as a viable strategy to enhance the yield of different crops and mitigate some of the current and future impacts of global warming and climate change on agriculture.

Physiological, biochemical and transcriptional responses to single and combined abiotic stress in stress-tolerant and stress-sensitive potato genotypes

Project description:In many potato cultivation regions, production is constrained by abiotic stresses such as drought and high temperatures which are often present in combination. We aimed to identify key mechanisms and processes underlying single and combined abiotic stress tolerance by a comparative analysis of tolerant and susceptible cultivars. Physiological data supported cultivars Desiree and Unica as being abiotic stress tolerant, while Agria and Russett Burbank were stress susceptible. This was indicated by the stronger impact of abiotic stress on photosynthetic carbon assimilation in the susceptible cultivars. Similarly, susceptible cultivars exhibited a lower leaf transpiration rate following stress, particularly combined heat and drought stress. Transcript profiles using microarrays were highly divergent both between genotypes and following the application of stress treatments. However, relatively few transcripts or metabolites exhibited genotype specific responses to abiotic stress treatment. Furthermore, apart from a decrease in the abundance of transcripts associated with PSII, particularly the light harvesting complex in both Desiree and Unica, there were very few changes that were consistent across stress susceptible or stress tolerant genotypes following stress treatment.

Gene expression of flower buds between rdr6-15 and wild-type under drought stress condition

Project description:To understand ta-siRNA-mediated regulation network in response to abiotic stress during flower development, gene expression profiles of flowering buds between rdr6-15 and wild-type under moderate drought stress and control condition were measured using microarray. 512 genes were bilaterally regulated by drought stress signal and RDR6-mediated signal. rdr6-15 and wild-type plants (Arabidopsis thaliana ecotype Columbia) were grown on 30g of vermiculite soil of plastic pot for 2 weeks. Then, water depletion (drought stress treatment) was applied for 1 week. Water content of the soil was decreased to less than 20%. 10 flower buds were collected for each experiment. The experiment was repeated 3-times.

ABAP1 is a negative regulator of the cell cycle that binds to transcription factors and represses their target gene expression, including pre-replication complex genes. All experiments were performed in triplicate.

To assess physiological effects of stress treatments, light responses (A/I) curves were measured on the third fully expanded leaf from four plants per treatment and in the control, immediately after heat stress, and when Î¨w was -0.9 MPa in water stress.
