Project description:Signaling between the cytokine TWEAK and its receptor Fn14 is critical for neural circuit development in the postnatal brain. However, the downstream mechanisms through which TWEAK and Fn14 mediate synapse development have remained unclear. Here, we apply single-nucleus RNA-sequencing to Fn14 and TWEAK KO mice to characterize the genes that are regulated by this pathway in the developing dorsal lateral geniculate nucleus of the thalamus at single-cell resolution. We identify robust cohorts of genes that are misregulated in the absence of either Fn14 or TWEAK, providing insight into the transcriptional mechanisms through which cytokine signaling between microglia and neurons regulates a key phase of postnatal brain development that is driven by sensory experience.

Microglia regulate Zika virus infection in human developing brain and glioma

Project description:This paper shows that, whereas SOX2+ cells in HDB are highly susceptible to ZKV, SOX2+ primary GBM samples exhibit moderate to high resistance to infection. These datasets address the reasons for this difference, based on infection of primary patient GBM and human developing brain samples with mCherry Zika reporter virus, then FACS sorting the Zika-mCherry positive and negative fractions from each sample for bulk RNA-Sequencing. The primary finding of importance is that Zika resistant GBMs exhibit an immune signature reflecting high levels of infiltration by microglia which are present only in small numbers or absent altogether in human developing brain.

Visual experience-dependent expression of Fn14 is required for retinogeniculate refinement (Fn14 KO RNA-Seq)

Project description:Sensory experience influences the establishment of neural connectivity through molecular mechanisms that remain unclear. Here, we employ single-nucleus RNA-sequencing to investigate the contribution of sensory-driven gene expression to synaptic refinement in the dorsal lateral geniculate nucleus of the thalamus, a region of the brain that processes visual information. We find that visual experience induces the expression of the cytokine receptor Fn14 in excitatory thalamocortical neurons. By combining electrophysiological and structural techniques, we show that Fn14 is dispensable for early phases of refinement mediated by spontaneous activity, but that Fn14 is essential for refinement during a later, experience-dependent period of development. Refinement deficits in mice lacking Fn14 are associated with functionally weaker and structurally smaller retinogeniculate inputs, indicating that Fn14 mediates both functional and anatomical rearrangements in response to sensory experience. These findings identify Fn14 as a molecular link between sensory-driven gene expression and vision-sensitive refinement in the brain.

TMT-coupled phosphoproteomics support novel role of TWEAK/Fn14 signaling in synaptic function

Project description:The study aims at illustrate novel role of TWEAK/Fn14 signaling in synapse function by combining electrophysiological and phosphoproteomic approaches. The results show that TWEAK acutely dampens basal synaptic transmission and plasticity through neuronal Fn14 and impacts the phosphorylation state of pre- and post-synaptic proteins in adult mouse hippocampal slices. Two models featuring synaptic deficits were used in the study. Blocking TWEAK/Fn14 signaling augments synaptic function in hippocampal slices from amyloid beta-overexpressing mice. After stroke, genetic or pharmacological inhibition of TWEAK/Fn14 signaling augments basal synaptic transmission and normalizes plasticity. Our data support a glial/neuronal axis that critically modifies synaptic physiology and pathophysiology in different contexts in the mature brain and may be a therapeutic target for improving neurophysiological outcomes.
