Proteomic response of Turicibacter bilis MMM721 to chicken bile and its bile acids

Project description:Bile and its individual components, mainly bile acids, are important for digestion and drive bacteria community dynamics in the upper gastrointestinal tract of chickens. However, specific responses to bile acids have been characterized in only a few commensal bacteria, and it is unclear how other members of the microbiota respond to biliary stress. Here, we used label-free LC-MS/MS to assess the proteomic response of a common inhabitant of the chicken upper intestinal tract, Turicibacter spp. MMM721, to 24 hours of growth in anaerobic growth media supplemented with 0.1% whole chicken bile, 0.1% taurochenodeoxycholic acid, or 0.1% taurocholic acid. 70, 46, and 8 differentially expressed proteins were identified in Turicibacter spp. MMM721 cultured with supplements of whole chicken bile, TCDCA, and TCA, respectively, when compared to unsupplemented controls. Many of the differentially expressed proteins were involved in ribosomal processes, post-translational modifications and chaperones, and modifications to the cell surface. To our knowledge, this work represents the first description of the Turicibacter spp. MMM721 proteomic response to bile and bile acid exposure. Ultimately, the T. bilis MMM721 response to whole bile and bile acids is highly complex, with numerous proteins from a variety of functional categories being induced.

Response of C. jejuni to various environmental conditions

Project description:Campylobacter jejuni is the leading cause of foodborne human gastroenteritis in the developed world. Infections are largely acquired from poultry produced for human consumption and poor food handling is thus a major risk factor. In this study, C. jejuni were exposed to growth in a number of enviornmental conditions representative of the human gastrointestinal tract, including 0.1% deoxycholate (DOC), under iron limitation (induced by 1 mM deferroxamine, in the presence of chicken 'juice' or 'exudate (the thaw water of frozen commerical chicken products) and in the presence of mammalian mucin.

Novel inducible filamentous sliding motility identified in pathogenic Bacillus cereus group species

Project description:We isolated an atmospheric contaminant, subsequently identified as a new strain of Bacillus mobilis, which showed a novel, robust, inducible filamentous sliding motility and completely colonized a bacterial culture plate in less than 48 h under some conditions. This flagella-independent sliding motility was characterized by long filamentous cells at the expanding edge, and was induced when cells were inoculated onto lawns of metabolically inactive Campylobacter jejuni cells, heat killed bacterial biomass, and milk or blood dried onto agar plates. Phosphatidylcholine (PC), bacterial membrane components, and sterile human fecal extracts were sufficient to induce filamentous expansion. Screening of eight other Bacillus spp. (five from the B. cereus group and three other Bacillus spp.) showed that filamentous motility was conserved amongst B. cereus group species to varying degrees. RNAseq of filamentously expanding cells collected from PC and milk lawn plates in comparison to rod-shaped cells from control plates revealed that genes related to metabolism, ion and amino acid transport were differently regulated, genes controlling sporulation were reduced, and some virulence genes (e.g., hblA/B/C/D and plcR) were increased. We hypothesize that the robust and conserved nature of filamentous motility in pathogenic B. cereus group species can enhance bacterial colonization during host colonization.
