Project description:In recent years there is a growing epidemiological indication of excess risk of cardiovascular disease at low doses of ionizing radiation without a clear-cut threshold. It is proposed that damage to the vascular endothelium is critical in radiation-related cardiovascular diseases. In order to identify and better understand the underlying molecular mechanisms of ionizing radiation on endothelial cells, we performed a microarray analysis on immortalized human coronary artery endothelial cells irradiated with different doses (0.00, 0.05, 0.10, 0.50 and 2.0 Gy). RNA was extracted at different time points after irradiation (1 day, 7 days, 14 days).

Project description:The cancer initiating cells (CICs) act as a tumor initiation source. Recent studies have shown that CICs contribute to chemoresistance and radioresistance. The aims of this study were to investigate the relationship of CD133+ liver CICs and radiation resistance and to define a possible mechanism for radioresistance in hepatocellular carcinoma (HCC). Total RNA is obtained from CD133 positive cells and CD133 negative cells at 0, 12 and 24 hours after radiation exposure.

Project description:Objective:Biomarkers of radiation injury are needed in planning therapeutic measures for cancer patients receiving radiation therapy and civilians exposed to nuclear events. Previous research has highlighted the impact of radiation damage, with cancer patients developing acute disorders including radiation induced pneumonitis or chronic disorders including pulmonary fibrosis months after radiation therapy ends. Discovery of biomarkers that predict these injuries will offer the potential to treat people proactively to mitigate this damage and improve quality of life. Recent research has highlighted the potential for messenger RNA (mRNA), microRNA (miRNA), and long non-coding RNA (lncRNA) to be used as radiation biomarkers. Our study focused on the changes in these RNAs at 48h after radiation exposure of mouse lung tissue to define biological pathway changes and determine potential biomarkers. Result: We observed sustained dysregulation of specific mRNAs, lncRNAs, and miRNAs across all doses. We observed gene dysregulation which can be used to develop both markers to identify no-exposure vs radiation exposure including Hba and Hbb mRNA which were dysregulated even at 1 Gy. We also observed genes which can indicate high dose exposure including Cpt1c and Pdk4. Gdf15, and Eda2r, mRNA markers of senescence and fibrosis, were the most significantly upregulated. Only three miRNAs were significantly dysregulated across all radiation doses, with miRNA-142-3p and miRNA-142-5p downregulated and miRNA-34a-5p upregulated. IPA analysis indicated that numerous pathways relevant to immune function, cell proliferation and survival decreased with increasing doses of radiation. This data highlighted early pathways of dysregulation depending on dose of exposure. This data will help with development of treatments and in medical decision-making. Further experiments are planned to develop medical countermeasures based on this early dysregulation.

Project description:Recent studies have further heightened awareness of the risk of radiation-induced cancer after diagnostic radiology imaging, in particular, brain cancer following childhood CT scans. One feature of Ptch1+/- mice is that they are sensitive to radiation-induced medulloblastomas (an embryonic cerebellar tumor) during a narrow time window centered on the days around birth. The dynamics of how dose-protraction interacts with such narrow windows of sensitivity in individual tissues is still unknown. Using medulloblastomas from irradiated Ptch1+/- mice on a hybrid C3H × C57BL/6 F1 background, we have previously shown that the alleles retained on chromosome 13 (which harbors the Ptch1 gene) reveal two major mechanisms of loss of the wildtype allele. The loss of parental alleles from the telomere extending up to or past the Ptch1 locus by recombination (spontaneous-type) accounts for almost all medulloblastomas in unirradiated mice, while tumors in irradiated mice often exhibited interstitial deletions which start downstream of the wildtype Ptch1 and extend up varying lengths towards the centromere (radiation-type). Here, Ptch1+/- mice were exposed to an acute dose of 0.1 or 0.5 Gy gamma rays in utero or postnatally, or the same radiation doses protracted over a 4-day period, and were monitored for medulloblastoma development. The results show dose-dependent and age-dependent induction of radiation-type tumors; that the size of the radiation-induced deletion differs with the dose-rate; and, that tumor latency may be related to the size of the deletion. The radiation signature allows for unique mechanistic insight into the action of radiation to induce DNA lesions with known causal relationship to a specific tumor type, particularly for doses and dose-rates more relevant to diagnostic radiology imaging and accidental exposure of populations to radiation.

Project description:Recent observations show that the single-cell response of p53 to ionizing radiation (IR) is “digital” in that it is the number of oscillations rather than the amplitude of p53 that shows dependence on the radiation dose. We present a model of this phenomenon. In our model, double-strand break (DSB) sites induced by IR interact with a limiting pool of DNA repair proteins, forming DSB–protein complexes at DNA damage foci. The persisting complexes are sensed by ataxia telangiectasia mutated (ATM), a protein kinase that activates p53 once it is phosphorylated by DNA damage. The ATM-sensing module switches on or off the downstream p53 oscillator, consisting of a feedback loop formed by p53 and its negative regulator, Mdm2. In agreement with experiments, our simulations show that by assuming stochasticity in the initial number of DSBs and the DNA repair process, p53 and Mdm2 exhibit a coordinated oscillatory dynamics upon IR stimulation in single cells, with a stochastic number of oscillations whose mean increases with IR dose. Project description:Introduction: Breast radiotherapy is currently “one size fits all” regardless of breast cancer subtype (eg. luminal, basal). However, recent clinical data suggests that radiation response may vary significantly among subtypes. Therefore, current practice leads to over- or under-treatment of women whose tumors are more or less radiation responsive. We hypothesized that this clinical variability may be due, in part, to differences in cellular radiation response. Methods: We exposed 16 biologically-diverse breast tumor cell lines to 0 or 5GY radiation. Microarray analysis was performed on RNA harvested from those cell lines. Samples were run in triplicate. Following quality assessment, differential gene expression analysis was performed using a two-way multiplicative linear mixed-effects model. A candidate radiation response biomarkers with biologically plausible role in radiation response, were identified and confirmed at the RNA and protein level with qPCR and Western blotting assays. Induction in human breast tumors was confirmed in 32 patients with paired pre- and post-radiation tumor samples using IHC and microarray analysis. Quantification of protein was performed in a blinded manner and included positive and negative controls.
