Transcriptomic and alternative splicing analyses reveal mechanisms of the difference in salt tolerance between barley and rice

Project description:Both barley (Hordeum vulgare) and rice (Oryza sativa) belong to Poaceae family, but differ greatly in salt tolerance. In order to understand molecular mechanisms in the difference of salt tolerance between the two species, the responses of transcriptomic profiles to salt stress were compared between rice (cultivar Nipponbare) and barley (accession XZ26) to reveal how alternative splicing (AS) coordinates with transcriptional regulation in adaptation to salt stress. Physiological study showed that XZ26 had higher salt tolerance than Nipponbare, as reflected by less growth inhibition, lower shoot Na+ concentration and higher K+/Na+ ratio when exposed to salt stress. Transcriptomic analysis showed that XZ26 had higher ROS scavenging ability, less degradation of protein kinases and enhanced anti-oxidation. Moreover, AS genes related to ion transporter genes and transcription factors could enhance and amplify K+/Na+ homeostasis and signal transduction cascades. We proposed that higher salt tolerance of barley accession XZ26 is attributed to its superior K+/Na+ homeostasis, tissue detoxication and less energy consumption. The present results provide insights at transcriptomic levels into reasons why barley has higher salt tolerance than rice.

Identification of non-coding RNA in sweet sorghum under salt stress

Project description:Salt stress has become one of the main abiotic stress factors restricting agricultural production worldwide. Sweet sorghum is an important salt and drought tolerant feed and energy crop. Its salt tolerance mechanism has not been widely studied. With the development of transcriptome sequencing technology, it is possible to study the molecular mechanism of sweet sorghum salt tolerance. The purpose of this study was to further reveal the potential salt-tolerant molecular mechanisms of sweet sorghum through high-throughput sequencing analysis of the transcriptome. Finally, through high-throughput sequencing, we read approximately 54.4G of raw base and 53.7G of clean base in total, and used FastQC to assign a quality score (Q) to each base in the read using a similar phred algorithm, Analysis shows that the data is highly credible. We conclude that RNA-based transcriptome characterization will accelerate the study of genetics and molecular biology of sweet sorghum salt tolerance mechanisms and provide a framework for this.

Quantitative Proteomics Analysis reveals Proteins related with high melatonin content in barley seeds under NaCl-induced salt stress

Project description:Increase soil salinization is affecting hull-less barley cultivation in the Qinghai-Tibet Plateau of China. Under saline conditions wild hull-less barley seeds showed high melatonin (MEL) accumulation during germination with improve salt tolerance. Hence, the present study aimed to understand the proteome level changes that result in high melatonin content in the germinating hull-less barley seed after treatment with 240mM-NaCl concentrations compared to control. Our results indicate that salt stress induced global changes in the proteome of germinating barley seeds. The NaCl treatment altered the expression and abundance of several proteins related to carbohydrate and energy metabolism, amino acid transport and metabolism which contributed to the high melatonin content in the seeds under salt stress. Further, proteins associated with cellular redox homeostasis, osmotic stress response and secondary metabolites derived from amino acid metabolism, including purine degradation, and shikimate pathways were induced in seeds with high melatonin content. Consequently, triggering a robust response to the oxidative stress occasioned by the NaCl-induced salt stress and lead to improved germination rate and salt stress adaptation in hull-less barley seeds. Overall our finding contribute to our understanding of the mechanism of melatonin mediated salt stress tolerance in barley at the protein level.
