Project description:We collected microsamples of the middle cerebral artery (MCA) from MMD patients (n = 21) and control patients (n = 11). Using microarray experiments, the profile of lncRNAs expression in MCA were compared between groups and differentially expressed lncRNAs (fold change > 2, q < 0.05) were identified. In addition, the neighboring coding gene whose transcription can be regulated in cis by the identified differentially expressed lncRNAs were investigated, and applied to Gene Ontology analysis for predicting those biological functions. We detected 308 differentially expressed lncRNAs (fold change > 2, q < 0.05), including 306 up-regulated and 2 down-regulated lncRNAs in the MCA from the MMD patients. As for the prediction of biological function, Gene Ontology analyses with possible coding genes whose transcription are regulated in cis by the identified differentially expressed lncRNAs suggested the involvement in antibacterial humoral response, T cell receptor signaling pathway, positive regulation of cytokine production, and branching involved in blood vessel morphogenesis. Quantitative polymerase chain reaction demonstrated a similar lncRNAs expression to the microarray experiments.

Project description:Moyamoya disease (MMD) is a cerebrovascular disease characterized by progressive stenosis of the intracranial internal carotid arteries and their proximal branches. However, the etiology of this rare disease remains widely unknown. Serum microRNA (miRNA) profiles have been screened to identify novel biomarkers for disease diagnosis and prognosis. Here, we identified important serum miRNAs that might play important roles in contributing to MMD pathogenesis through microarray analysis. 10 MMD patients and 10 controls were consecutively recruited at Shanghai Changhai Hospital. Five-ml venous blood was collected from each participant and separated into serum and cellular fractions. We pooled serum samples from 10 MMD patients and 10 controls. Agilent Human 8 x 60K miRNA Array was performed on the two pooled samples.

Epigenome analysis of intracranial aneurysm and superficial temporal artery samples

Project description:Genomic DNA of intracranial aneurysm and matched superficial temporal artery from same patient (n=9) was extracted with QIAamp DNA Mini Kit (Qiagen, Valencia, CA). A total of 500 of genomic DNA was bisulfite conversion using the EZ DNA Methylation Gold Kit (Zymo Research, Irvine, CA) and then analyzed on an Infinium HumanMethylation450 BeadChip (Illumina, San Diego, CA) following the manufacturer’s instructions.

RNA difference in superficial temporal artery between moyamoya disease and normal control

Project description:We collected the Superficial temporal artery (STA) tissues from patients with Moyamoya disease who underwent combined direct and indirect bypass surgery and patients with brain trauma requiring craniotomy in the Department of Neurosurgery, First Affiliated Hospital of USTC (Anhui Provincial Hospital). One part was fixed in 10% neutral formalin solution, and the other part was stored in a refrigerator at -80 ℃. All protocols using human specimens were approved by the ethics committee of the First Affiliated Hospital of USTC (Anhui Provincial Hospital). Written informed consent was obtained from all patients. All protocols were approved by the Institutional Review Board of the First Affiliated Hospital of USTC (Anhui Provincial Hospital).Total RNA was extracted from the STA tissues using Trizol reagent (Invitrogen, Carlsbad, CA, USA) according to the manufacturer's protocol. The integrity and concentration of RNA were detected using an Agilent Bioanalyzer 2100 system (Agilent Technologies, Santa Clara, Calif., USA), enriched and purified with Oligo (dT) -bearing magnetic beads. RNA sequencing was performed by Anoroad (Beijing, China).
