Project description:miRNA expression profiling of Pinellia pedatisecta leaves comparing Pinellia ternata leaves in the same condition. Then we compared the accumulation of known miRNA families using microarray signals to identify further the differential miRNA expression between Pinellia pedatisecta and Pinellia ternata. Pinellia pedatisecta vs Pinellia ternata, independently grown and harvested. Each sample are mixed. One replicate per array.

Large-Scale Analysis of Protein Crotonylation Reveals Its Diverse Functions in Pinellia ternate

Project description:We generated a global crotonylome analysis of the leaves in Pinellia ternata. We found 4509 crotonylated sites on 1757 proteins totally, and three specific amino acids surrounding crotonylation sites were identified in Pinellia ternata: KcrF, K***Y**Kcr and Kcr****R. Gene Ontology (GO) and KEGG pathway enrichment analyses showed that two crucial alkaloid biosynthesis related enzymes and many stress-related proteins were also highly crotonylated. Furthermore, several enzymes participate in the carbohydrate metabolism pathway, were modified also by lysine crotonylation.

Phenotypic and Transcriptomic Analysis of Two Pinellia ternata Varieties T2 line and T2Plus line

Project description:Pinellia (Pinellia ternata (Thunb.) Breit.), as important medicinal plant, has been used to treat various ailments for a long time. The sixteen ploid plant (2n=16*13=208) Pinellia T2Plus line was obtained from an octoploid (2n=8*13=104) T2 line by chromosome-doubling technique. Compared with T2 line, the content of various medicinal components (polysaccharide, guanosine, adenosine and ephedrine) was increased in T2Plus line. In this study, the transcriptome of T2 line and T2Plus line were characterized by RNA sequencing (RNA-seq) technology. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways enrichment analysis on differential expressed unigenes (DEGs) revealed that multiple metabolic pathway were enriched significantly, such as ‘Starch and sucrose metabolism’, ‘Purine metabolism’, ‘Photosynthesis’ and six transcription factors (MYB, WRKY, bHLH, LBD lateral organ boundaries domain (LBD), HD-ZIP homeodomain-zipper (HD-ZIP) and ERF Ethylene-responsive factor (ERF) ) play a key role in difference of transcriptome between T2 line and T2Plus line. These metabolic pathways and transcription factors may play an important role in the difference of medicinal components and epigenetic features between these two Pinellia cultivars. This conclusion provides a robust theoretical basis for the mechanism of the formation of medicinal ingredients in Pinellia cultivars.

Integrated analysis of transcriptome and miRNAome reveals the heat stress response of Pinellia ternata seedlings

Project description:Pinellia ternata (Thunb.) Briet., a valuable herb native to China, is susceptible to the "sprout tumble" phenomenon due to high temperatures, resulting in a significant reduction in yield. The molecular regulatory mechanisms underlying the response of P. ternata to heat stress are not well understood. In this study, we integrated transcriptome and miRNAome sequencing to identify heat response genes, microRNAs (miRNAs), and key miRNA-target pairs in P. ternata under heat stress and room temperature conditions. Transcriptome analysis revealed extensive reprogramming of 4,960 genes across various categories, predominantly associated with cellular and metabolic processes, responses to stimuli, biological regulation, cell parts, organelles, membranes, catalytic and binding activity. miRNAome sequencing identified 1,597 known/conserved miRNAs that were differentially expressed in two test conditions. According to the analysis, genes and miRNAs associated with regulation of transcription, DNA-templated, transcription factor activity and sequence-specific DNA binding pathway, may play a major role in the resistance to heat stress in P. ternata. Integration of expression data of transcriptome and miRNAome determined high confidence 41 miRNA-mRNA pairs forming 25 modules. MYB-like protein and calcium-responsive transcription coactivator may play an integral role in the resistance to heat stress in P. ternata. In addition, the candidate genes and miRNAs were subjected to quantitative real-time PCR to evaluate their expression patterns. These results offer a foundation for future studies exploring the mechanism and critical genes of resistance to heat stress in P. ternata.
