Project description:To investigate the molecular mechanism of LKB1 in the regulation of lung development, we performed single-cell RNA-seq analysis of the lungs of Nkx2.1 Cre;Lkb1f/f and Lkb1f/f mice. In total, 30,408 lung cells were collected for the scRNA-seq analysis of the whole lung cells isolated from 5-week-old Nkx2.1 Cre;Lkb1f/f or Lkb1f/f mice, when the Nkx2.1 Cre;Lkb1f/f mice began to die.

Project description:The experiment was performed to find selective targets of autophagy in different subtypes of neurons. Therefore, brain tissue of conditional knockout mice lacking autophagy protein ATG5 in inhibitory or excitatory neurons was used. Cortices (ATG5flox:CamKII-Cre) / striata (ATG5flox:VGAT-Cre) of mice were mixed with lys6 labeled brain lysates as internal controls between the samples. Samples were measured by LC-MS/MS and resulting H/L ratios were used to quantify proteomic and phosphoproteomic changes.

Project description:We employed genetic mouse model (Fsp1-Cre;Col1a1flox/flox) to delete Col1a1 (type I collagen) specifically in Fsp1-expressing cells in comparison with control WT mice (Cre-negative;Col1a1flox/flox). Single-cell RNA-sequencing analyses were performed on unfractionated live cell mixtures from bone marrow samples from Fsp1-Cre;Col1a1flox/flox mice and WT mice, to investigate the impact of Fsp1-cell-specific Col1 deletion on bone marrow microenvironment.
