Project description:Plant volatiles can mediate plant-plant communication in the sense that plants attacked by herbivores can signal their unattacked neighbors of danger by emitting HIPVs. We call this the priming effect. Since the plant defense response is a systematic process involving numerous pathways and genes,to characterize the priming process, a time course study using a genome-wide microarray may provide more accurate information about the priming process. Furthermore, to what extent do the priming process and direct defense share similar gene expression profiles or pathways are also not clear. We used microarray to detect the priming effect of plant volatiles to healthy Arabidopsis thaliana, and the effect of direct leafminer feeding to Arabidopsis thalianas.

A system using Lima bean plants, from which HIPVs can be effectively induced by leafminer feeding, as emitters and Arabidopsis thaliana as receivers is used to track the priming process between neighbor plants. The Arabisopsis thaliana seedlings were treated by volatiles from leafminer fed lima bean for 24h or 48h for RNA extraction and hybridization on Affymetrix microarrays. The Arabisopsis thaliana seedlings fed by leafminer directly were also collected The for RNA extraction and hybridization on Affymetrix micorarrays. We want to explore the response of Arabidopsis thaliana to priming volatiles during a 24h-48h time course. We also want to compare the effect of priming and direct leafminer feeding.

Project description:Key Message Powdery mildew resistance in two strawberry mapping populations is controlled by both stable and transient novel QTL of moderate effect. Some low transferability of QTL across wider germplasm was observed. The obligate biotrophic fungus Podosphaera aphanis is the causative agent of powdery mildew on cultivated strawberry (Fragaria?×?ananassa). Genotypes from two bi-parental mapping populations 'Emily'?×?'Fenella' and 'Redgauntlet'?×?'Hapil' were phenotyped for powdery mildew disease severity in a series of field trials. Here, we report multiple QTL associated with resistance to powdery mildew, identified in ten phenotyping events conducted across different years and locations. Six QTL show a level of stable resistance across multiple phenotyping events; however, many other QTL were represented in a single phenotyping event and therefore must be considered transient. Subsequent screening of identified QTL across a validation set determined whether identified QTL remained closely linked to the associated resistance gene in the wider germplasm. Furthermore, a preliminary association analysis identified a novel conserved locus for further investigation. Our data suggest that resistance is highly complex and that multiple, primarily additive, sources of quantitative resistance to powdery mildew exist across strawberry germplasm. Utilisation of the reported markers in marker-assisted breeding or genomic selection would lead to improved powdery mildew-resistant strawberry cultivars, particularly where the studied parents, progeny and close pedigree material are included in breeding germplasm.

Project description:WRKY transcription factors play a nonnegligible role in plant growth and development, but little is known about the involvement of WRKY transcription factors in the regulation of fruit ripening. In this study, FaWRKY71 was identified to be closely related to fruit maturation in octoploid strawberry. FaWRKY71 protein localized in the nucleus and responded to cold, salt, low phosphate, ABA, and light quality in strawberry seedlings. The temporal and spatial pattern expression analysis indicated that FaWRKY71 was expressed in all the detected tissues, especially in the full red fruits. In addition, FaWRKY71 gave rise to the accumulation of anthocyanin content by promoting the expression of structural genes FaF3'H, FaLAR, FaANR, and transport factors FaTT19 and FaTT12 in the flavonoid pathway, and softening the texture of strawberry via up-regulating the abundance of FaPG19 and FaPG21. Furthermore, FaWRKY71 was a positive regulator that mediated resistance against reactive oxygen species by enhancing the enzyme activities of SOD, POD, and CAT, reducing the amount of MDA. Altogether, this study provides new and comprehensive insight into the regulatory mechanisms facilitating fruit ripening in strawberry.
