Project description:Circular RNAs (circRNAs) are key regulators in the development and progression of human cancers. A human circRNA microarray analysis was performed to screen for abnormally expressed circRNAs in breast cancer tissue and circRNA-000911 was identified as a circRNA which was significantly downregulated in breast cancer cells. Mechanistic investigations suggested that the enhanced expression of circRNA-000911 suppressed cell proliferation, migration and invasion, and promoted the apoptosis of breast cancer cells. The overexpression of circRNA-000911 may provide a future direction which may aid in the development of a novel treatment strategy for breast cancer.
Project description:Arraystar Human circRNA Microarray is designed for the global profiling of human circRNAs. In this study, we applied a circRNA microarray to screen the potential biomarker for HCC. 20 samples extracted from plasma samples including HCC group before operation, and after operation, CH group and control group. Each group contained five samples.
Project description:We collected 3 paired tumor tissues and adjacent normal tissues collected from patients with node-positive primary breast cancer. Total RNA was extracted from tissues using Trizol reagent. Then total RNA was processed and circRNAs were profiled using RNA-seq technique. Raw read counts were filtered to remove the genes with a zero read count in 3 or more samples, and then normalized with DESeq2 in the statistical program R (version 4.0.1). Differentially expressed genes were screened using following criterions: fold change was larger than 2 and adjusted P value was less than 0.05.
