Project description:Knee ligaments, the anterior cruciate ligament (ACL) and medial collateral ligament (MCL), have unique functions but distinct regeneration capacities. However, the landscape of cellular heterogeneity in ligaments remains unclear. Here we profile the transcriptional signatures of ACL and MCL cells using single-cell RNA-sequencing. These ligaments consist of three fibroblast types that express Col22a1, Col12a1, or Col14a1 but have distinct localizations in the tissue. In addition, we find substantial heterogeneity in Col12a1 and Col14a1 positive cell clusters when comparing the ACL and MCL. Furthermore, we identified aging related changes in both cell composition and gene expression in ligaments.

Single-cell sequencing of murine articular cartilage

Project description:Osteoarthritis (OA) is a degenerative joint disease characterized by progressive cartilage loss, bone remodeling, synovial inflammation, and significant joint pain, often resulting in disability. Injury to the synovial joint such as the anterior cruciate ligament (ACL) tear is the major cause of OA in young adults. Currently, there are no approved therapies available to prevent joint degeneration or rebuild articular cartilage destroyed by OA, primarily because our understanding of the cellular and molecular changes that contribute to joint damage is very limited. The synovial joint is a complex structure composed of several tissues including articular cartilage, subchondral bone, synovium, synovial fluid, and tensile tissues including tendons and ligaments. In the present study, using single-cell RNA sequencing (scRNA-seq), we examined the cellular heterogeneity in articular cartilage from mouse knee joints and determined the knee joint injury-induced early molecular changes in the chondrocytes that could contribute to OA.

Lgr5 and Col22a1 mark progenitor cells in the lineage toward juvenile articular chondrocytes

Project description:The synovial joint forms from a pool of progenitor cells in the interzone region of the future joint. Expression of Gdf5 and Wnt9a has been used to mark the earliest cellular processes in the formation of the interzone and the progenitor cells. However, progression and lineage specification toward the different tissues of the joint is not well understood. Here, by lineage tracing studies we identify a population of Lgr5+ interzone cells that contribute to the formation of cruciate ligaments, synovial membrane and articular chondrocytes of the joint. This is supported by single cell transcriptome analyses. Furthermore, we showed that Col22a1, a marker of early articular chondrocytes, co-expresses with Lgr5+ cells prior to cavitation as an important lineage marker specifying the progression towards articular chondrocytes. Lgr5+ cells contribute to the repair of a joint defect with the re-establishment of Col22a1 containing superficial layer.

Single-cell transcriptome analysis reveals cellular heterogeneity in mouse intra- and extra articular ligaments.

Project description:Ligaments are collagenous connective tissues that connect bones. Injury of knee ligaments, namely anterior cruciate ligament (ACL) and medial collateral ligament (MCL), is common in athletes. Both ligaments have important functions, but distinct regeneration capacities. The capacity for recovery after injury also diminishes with age. However, cellular heterogeneity in the ligaments remains unclear. Here, we profiled the transcriptional signatures of ACL and MCL cells in mice using single-cell RNA sequencing. These ligaments comprise three fibroblast types expressing Col22a1, Col12a1, or Col14a1, but have distinct localizations in the tissue. We found substantial heterogeneity in Col12a1- and Col14a1-positive cells between ACL and MCL. Gene Ontology analysis revealed that angiogenesis- and collagen regulation-related genes were specifically enriched in MCL cells. Furthermore, we identified age-related changes in cell composition and gene expression in the ligaments. This study delineates cellular heterogeneity in ligaments, serving as a foundation for identifying potential therapeutic targets for ligament injuries.
