Project description:In the present study we aimed to investigate whether alterations in left ventricular myocardial micro-RNA (miRNA) expression and post-transcriptional gene regulation might contribute to the development of systolic heart failure in aortic-banded (AB) rats. Hence, in young adult male Sprague-Dawley rats abdominal AB was carried out. In 10% of the AB animals, congestive systolic heart failure developed after 12 weeks of follow-up. These rats made up the AB-heart failure group. On the other hand, in the majority of the AB rats (almost 90%), heart failure signs were not present. These rats made up the AB-hypertrophy group. Additionally, sham-operated animals served as controls. miRNA profiling was conducted in all the three experimental groups. Based on the NGS data, network theoretical miRNA-target analysis was than performed to predict target genes. The mRNA expression of the predicted genes were finally measured by qPCR.

Genome-wide maps of H3K9me3 and H3K4me3 state in failing heart with or without chaetocin in animal model

Project description:Epigenetic status has been linked to cardiac hypertrophy and heart failure. Histone deacetylase inhibitors are promising drugs for preventing cardiac remodeling. We previously demonstrated very different patterns of histone H3 lysine 9 trimethylation (H3K9me3) and histone H3 lysine 4 trimethylation (H3K4me3) in failing hearts compared to control hearts in both animal models and clinical heart specimens. Here, we focused on a heart failure-specific histone modification, H3K9me3, and investigated the prognostic efficacy of administering a histone H3K9 methyltransferase inhibitor, chaetocin, to Dahl salt-sensitive rats, an animal model of heart failure. Chaetocin delayed the timing of transition from cardiac hypertrophy to heart failure, and prolonged survival in this animal model. Mitochondrial dysfunction was improved with inhibitor use in the failing heart. ChIP-seq analysis demonstrated that heart failure caused an increase in H3K9me3 alignments in thousands of repetitive elements, including regions neighboring mitochondrial genes, and a corresponding reduction of this effect with inhibitor use. However, at 35 loci, heart failure was conversely associated with a reduction in H3K9me3 alignments, and inhibitor use reversed this effect. These data suggest that excessive heterochromatinization of repetitive elements in the failing heart might impair pumping function with mitochondrial gene silencing. H3K9 methyltransferase inhibitors may be a promising novel therapy for chronic heart failure.

Transcription profiling of rat colon after dietary treatment with fructo oligosaccharides

Project description:To increase our knowledge of the effects of Fructo oligosaccharides (FOS) on the intestinal barrier function in rats, a controlled rat infection study was performed. Two groups of rats were adapted to a diet with or without FOS. mRNA was collected from the mucosa of the colon and changes in gene expression were assessed using an agilent rat whole genome microarray (G4131A Agilent Technologies). Results indicate that dietary FOS influences energy metabolism, which will most likely play a role in the effects of FOS on the intestinal barrier. Experiment Overall Design: In the present study, large-scale gene expression analysis was performed to reveal mechanistic details of FOS induced gene expression in vivo in the colon mucosa. Wistar rats were adapted to diets with or without FOS for 14 days. RNA was isolated from colonic mucosal scrapings. Agilent rat whole genome microarray containing 44290 60-mer spots, were used to study FOS induced gene expression changes in order to better understand the FOS induced effects on the intestinal barrier of rats.
