Project description:In the present study, an iTRAQ-based quantitative proteomic analysis, combined with gene expression and virus-induced gene silencing (VIGS), were used to identify genes associated with the infection of kiwifruit (Actinidia deliciosa ‘Hayward’) by B. cinerea. To the best of our knowledge, this is the first proteomic study of the kiwifruit-B. cinerea interaction, and provides information that can be used to better understand the mechanism of gray mold infection in kiwifruit.

Project description:Inflammatory Bowel Disease (IBD) is a term describing a collection of conditions characterised by chronic inflammatory disorder of the gastrointestinal tract involving an inappropriate immune response to commensal microorganisms in a genetically susceptible host. Four kiwifruit extracts, aqueous and ethyl acetate extracts of gold kiwifruit (Actinidia chinensis) or green kiwifruit (A. deliciosa), have previously demonstrated anti-inflammatory activity using in vitro models of IBD. This study examined whether these kiwifruit extracts had immune modulating effects in vivo against inflammatory processes known to be increased in patients with IBD. KFEs were used as a dietary intervention in Il10-/- mice (an in vivo model of IBD) and the C57BL/6J background strain in a 3 x 2 factorial design. While all Il10-/- mice developed significant colonic inflammation compared to the C57BL/6J mice, this was not affected by the inclusion of KFE in the diet. Whole genome gene and protein expression level profiling indicated that KFEs influenced immune signalling pathways and metabolic processes within the colonic tissue; however, the effects were subtle. In particular, adaptive immune pathways were reduced by three out of four kiwifruit extracts, with greater reduction seen in the C57BL/6J mice. This suggests that while immune-modulating activity was present in vivo, KFEs did not reduce inflammatory processes relevant to IBD. Experimental design. Two experiments were conducted, one using extracts from gold kiwifruit and one using extracts from green kiwifruit. Project description:The response of kiwifruit vine (Actinidia chinensis 'Hort16A') canes to feeding by the armoured scale insect Hemiberlesia lataniae was tested by screening RNA extracted from bark against a cDNA library of 17,512 unigenes. Substantial transcriptional changes were noted after 7 days for approximately 30% of the ESTs. Transcripts associated with photosynthesis were down-regulated and those associated with secondary metabolism up-regulated. A large number of transcripts orthologous with defence-related genes were differentially expressed including several orthologous with resistance and pathogenesis. This is the first such analysis of changes to transcript expression in a plant challenged by feeding by insects from the family diaspididae.

Project description:To study the responses of kiwifruit to Pseudomonas syringae pv. actinidiae, one-year-old potted seeding A. c. var. deliciosa cultivar ‘Jinkui’ and the pandemic Pseudomonas syringae pv. actinidiae bacterial strain JF8 (CCTCC AB2018305) were used for this study. This bacterial strain was originally isolated from A. c. var. chinensis cultivar ‘Jinfeng’ and further characterized . Plants were maintained in an aseptic room, with 95% of relative humidity, have natural light and no further fertilization after their receiving from the nursery. For inoculation, the P. s.pv. actinidiae strain was streaked on nutrient-sucrose agar (NSA) and incubated at 25 °C for 48-h. Ten microliters of a bacterial suspension (1-2×107cfu/mL) prepared in sterile 0.85 % w NaCl were inoculated in the plants chosen for investigation. The bacterial suspension was sprayed to entirety tree. In parallel, control plants were treated in the same way with sterile 0.85 % w NaCl solution. The inoculated and control plants were randomly distributed in the room at 15 ± 3 °C. 24-h after inoculation, ‘Jinkui’ leaves were sampled from the infected and control plants for further analyses. Each sample consisted of the leaves of one tree. Three biological replicates were used for each line.
