Protein profile of commercial rice milks analyzed by label-free quantitative proteomics

Project description:Rice milk is a non-diary beverage made primarily from milled rice and water that has an opaque white or beige color and creamy texture resembling that of cow’s milk. Rice milk is generally considered hypoallergic and gluten-free hence, is an excellent choice for people with lactose intolerance and multiple allergies. Besides having anti-inflammatory properties, it is beneficial in lowering cholesterol and hypertenstion and is known to contain a range of bio-active components such as α-tocopherol, c-oryzano, ß-sitosterol, thiamin, niacin and pyridoxine. The consumption of rice milk is increasing worldwide for its nutritional value and health benefits, however, its protein composition after commercialization is not well known. This study compared the protein profile of 9 different commercial rice milks using a label-free quantitative proteomics approach.

Project description:Milk protein is one of the most important economic traits in the dairy industry. Yet, the miRNA gene regulatory network for the synthesis of milk protein in mammary is poorly understood. In this study, the hypothesis was that miRNAs have potential roles in bovine milk protein production. Using miRNA-seq and RNA-seq, we investigated the miRNAs profiles of mammary glands from 12 Chinese Holstein cows with six cows at peak of lactation and six in non-lactating period, from which three cows were in high and three in low milk protein percentage.

Genomic characterization of normal and aberrant human milk production [scRNA-seq]

Project description:Breastfeeding is vital for reducing morbidity and mortality, yet exclusive breastfeeding rates are low, with insufficient milk supply being a major weaning factor whose molecular causes remain largely unknown. In this study, we collected fresh milk samples from 30 lactating individuals, classified as low, normal, or high milk producers at multiple postpartum stages, and conducted extensive genomic and microbiome analysis. Using bulk RNA sequencing on human milk fat globules (MFG), milk cells, and breast tissue, we found that MFG-derived RNA closely resembles RNA from milk luminal cells. Furthermore, bulk and single-cell RNA-seq revealed changes in the transcriptome and cellular content linked to milk production. We identified specific genes and cell-type proportions differing in low and high milk production. Infant microbiome diversity was affected by feeding type, but not by maternal milk supply. This study provides a comprehensive human milk transcriptomic catalog, identifies genes associated with milk production, and highlights MFG as a useful biomarker for milk transcriptome analysis.
