Project description:Rainbow darter (Etheostoma caeruleum) are a small benthic fish found in North America. This species is sensitive to sewage effluent in the environment, showing the presence of intersex in up to 80% of males in near-field areas in the Grand River, ON. To learn more about the molecular events associated with intersex, we developed a customized oligonucleotide microarray (4x180K) with next generation sequencing (454 Roche) to characterize molecular responses in the gonad. Transcriptomics was performed on both males and females from both a reference site and a polluted site. Males with and without intersex from the polluted site were compared to the control males. Rainbow darter were sampled from from the Grand River in May 2011. Fish were selected according to the location, gonad maturity, and intersex index. Reference fish were taken from the upstream to the urban area; exposed fish were taken from downstream of from Kitchener MWWE treatment plant.

Project description:This repository contains whole genome long read sequencing data generated using Oxford Nanopore Technologies from a mouse of the Four Core Genotypes cross. By crossing mice with both a deletion of the sex-determining factor Sry on the Y-chromosome and a transgenic insertion of Sry on Chromosome 3, four combinations of gonadal (testis or ovaries) and chromosomal (XX or XY) are generated, namely XYSry-Chr3Sry+ (gonadal and chromosomal males), XYSry- (gonadal females, chromosomal males), XX (gonadal and chromosomal females), XXChr3Sry+ (gonadal males, chromosomal females). The transgenes were on a C57BL6 genetic background which is crossed with a CAST/EiJ female to allow for the distinction of the parental haplotypes. DNA sequencing was done on a liver sample of the XYSry+ genotype.

Project description:Ovotestis often occurs in intersex individuals under certain pathological and physiological conditions. However, how ovotestis is formed remains unknown. Here, we report the first comprehensive single-cell developmental atlas of the model fish ovotestis. We provide an overview of ovotestis cell identities and a roadmap of germline, niche, and stem cell development in ovotestis by cell lineage reconstruction and a uniform manifold approximation and projection. We identify common progenitors of germline stem cells with two states, which reveal their bipotential nature to differentiate into both spermatogonial stem cells and female germline stem cells. Moreover, we found that ovotestis infertility was caused by developmental defects in oogenesis owing to dynamic autophagic degradation in female germline cells, and in spermatogenesis due to deficiency of histone-to-protamine replacement in spermatid differentiation. Notably, signaling pathways in gonadal niche cells and their interaction with germline cells synergistically determined cell fates of both male and female germlines in ovotestis. Overall, we reveal a cellular fate map of germline and niche cell development that shapes cell differentiation directions of ovotestis, and provide novel insights into ovotestis development.

Project description:Environmental estrogens are ubiquitous in aquatic systems worldwide. In wild fish inhabiting wastewater effluent with estrogenic components, the presence of both oocytes and spermatogenic tissue in the testes has been documented. However, the molecular networks underlying the manifestation of the intersex condition are not completely characterized. In this study, we used an environmentally relevant concentration of 17alpha-ethinylestradiol (EE2), the active component of the birth control pill, to determine the response of the fathead minnow testis transcriptome after 96 hrs. The goal was to capture early molecular initiating events that may lead to the intersex condition following exposure to an estrogen that is present in wastewater effluent. There were no effects of EE2 on morphometric endpoints such as gonadosomatic index nor proportion of gametes within the testes. However, in vitro production of 11-ketotestosterone and testosterone in the testis was decreased relative to controls following EE2 exposure. Gene expression profiling revealed that there were 10 transcripts that were differentially expressed in the testis using a 8x60K fathead minnow microarray, the most dramatic change being that of coagulation factor XIII A chain (20-fold increase). Transcripts that included guanine nucleotide binding protein (Beta Polypeptide 2), peroxisome proliferator-activated receptor delta, and WNK lysine deficient protein kinase 1a, were down-regulated by EE2. Subnetwork enrichment analysis revealed that EE2 suppressed transcriptional networks associated with reproduction such as steroid metabolism, hormone biosynthesis, and sperm mobility. Most interesting was that gene networks associated with doublesex and mab-3 related transcription factor 1 (dmrt1) were suppressed in the adult testis, despite the fact that dnmt1 itself was not differentially expressed from controls. Moreover cell processes involving forkhead box L2 (foxL2) such as ovarian follicle development and granulosa cell development were increased in expression in the testis. Cell processes that included glucose homeostasis, response to heavy metal, amino acid catabolism, and the cyclooxygenase pathway were decreased with EE2 while lymphocyte chemotaxis, intermediate filament polymerization, glucocorticoid metabolism, carbohydrate utilization, and anterior/posterior axis specification were increased, shedding light on the transcriptional responses that may be perturbed prior to gonadal remodelling following exposure to estrogenic contaminants. These data demonstrate that low concentrations of EE2 rapidly suppresses male hormone secretion, suppresses gene networks related to male sex differentiation, and induces transcriptional networks related to female sexual differentiation in the adult testis. These responses are hypothesized to be the molecular initiating events that occur prior to the development of the intersex phenotype after prolonged exposures to estrogens.
