Project description:The emerging evidences support that exosome cargo miRNAs function as important regulators in cell differentiation. Therefore, in order to figure out the mechanism that Exo-AT mediated adipogenesis, we profiled miRNAs in Exo-AT using high-throughput sequencing (miRNA-seq). After trimming low-quality reads, contaminants, adaptors, and reads smaller than 15 nt, the remaining reads were mapped to merged pre-miRNA data bases. To identify the conserved miRNAs in Exo exosomes, miRNAs were aligned to miRBase v21. 148 and 154 types of known miRNAs in Exo-ADSCs and Exo-AT, respectively, were identified in the two replicates. Among these miRNAs, 103 miRNAs were simultaneously detected in both Exo-ADSCs and Exo-AT. Compared to Exo-ADSCs, 45 conserved miRNAs were enriched (expressed ≥ 2 folds, FDR<0.05) in Exo-AT. KEGG Pathway analysis was performed for the targets of the most 20 enriched miRNAs in Exo-AT (compared with Exo-ADSCs) to determine their potential function. Data showed that pathways that regulate adipogenesis such as Wnt signaling pathway, Insulin signaling pathway, MAPK signaling pathway, TGF-ß signaling pathway were enriched significantly for targets of Exo-AT miRNAs. Furthermore, 14 of 45 enriched miRNAs in Exo-AT (31.11%, such as miR-30a-5p, miR-148a-3p) were reported to participate in regulation of adipogenesis while 8 miRNAs (17.78%, such as miR-93-5p, miR-150-3p) that negatively control osteoblastic differentiation of MSC have been described.

Project description:High-fat diet (HFD)-induced obesity is a multi-factorial disease including genetic, physiological, behavioral, and environmental components. Drosophila has emerged as an effective metabolic disease model. Cytidine 5'-triphosphate synthase (CTPS) is a crucial enzyme for the de novo synthesis of CTP, governing the cellular level of CTP and phospholipid synthesis. CTPS has been found to form filaments known as cytoophidia, which are evolutionarily conserved in bacteria, archaea, and eukaryotes. Here, we show that CTPS functions in fat bodies to regulate body weight and starvation resistance in Drosophila. HFD-induced obesity enhances CTPS transcription and lengthens cytoophidia in larval adipocytes. CTPS depletion in the fat body prevented HFD-induced obesity, including body weight gain, adipocyte expansion, and lipid accumulation, by inhibiting the PI3K-Akt-SREBP axis. A dominant-negative form of CTPS also inhibits adipocyte expansion and down-regulates lipogenic genes. As a result, our findings not only establish a functional link between CTPS and lipid homeostasis but also highlight a potential role of CTPS manipulation in the treatment of HFD-induced obesity.

Project description:Background:The objective of this study was to investigate obesity-related glomerulopathy (ORG) at a cellular, structural and transcriptomic level. Methods:Thirty Wistar rats were randomized into two groups: control rats(n=15), which were fed a standard diet(SD) and study rats(n=15), which were fed a high-fat diet (HFD). After 10 weeks, weight, parameters of kidney function, renal histological features, transcriptomic changes, miRNA and mRNA isolation were compared. Results:HFD gained more weight(55.8%) than SD(29.2%), p<0.001. Albuminuria was also significantly higher in HFD(10,384.04 ng/ml) compared with SD(5,845.45 ng/ml), p<0.001. HFD showed typical lesions of early stages of ORG, with a predominance of mesangial matrix increase (MMI) and podocyte hypertrophy (PH). All histologic lesions correlated with genes differentially expressed (DE) in kidneys of HFD group and PH, also correlated with specific miRNAs DE in the urine of HFD. The functional analysis showed 4 miRNAs DE in the kidneys of HFD group that negatively regulates PTEN gene, which promotes podocyte endocytosis of lipids in ORG. The electronic microscope confirmed the spaces of lipid vacuoles in the podocytes of HFD. Between those 4 miRNAs DE, miR-205 was also found to be upregulated in the urine of HFD group. Conclusions: Wistar rats fed a HFD developed early-stages of ORG, with a specific targetome of miRNAs and gene expression. The upregulation of miR-205 in kidney and its isolation in urine is associated with lipid endocytosis of podocytes, which could become a plausible biomarker of early-stages of ORG and open new avenues for future therapeutics research. Translational Statement: The findings of the basic research in this study can be replicated in human models of obesity. The results of the study on miRNA in humans may contribute to improving the understanding of the pathophysiology of obesity-related glomerulopathy, aiding in the search for early biomarkers, and exploring new therapeutic targets. Project description:There is an increasing clinical evidence that obesity exerts deleterious effects on the skeleton. While obesity coexists with estrogen deficiency in postmenopausal women, their combined effects on the skeleton are poorly studied. Thus, we investigated the impact of high-fat diet (HFD) on bone and metabolism of ovariectomized (OVX) female mice (C57BL/6J). OVX or sham operated mice were fed either HFD (60%fat) or normal diet (ND) (10%fat) for 12 weeks. HFD-OVX group exhibited pronounced increase in body weight (~86% in HFD and ~122% in HFD-OVX, p<0.0005) and impaired glucose tolerance. Bone microCT-scanning revealed a pronounced decrease in trabecular bone volume/total volume (BV/TV) in HFD-OVX (15.6±0.48% in HFD and 37.5±0.235% in HFD-OVX, p<0.005) and expansion of bone marrow adipose tissue (BMAT) (+60.7±9.9% in HFD vs +79.5±5.86% in HFD-OVX, p<0.005). Mechanistically, HFD-OVX treatment led to upregulation of genes markers of senescence, bone resorption, adipogenesis and inflammation and downregulation of gene markers of bone formation and bone development. Similarly, HFD- OVX treatment resulted in significant changes in bone tissue levels of Purine/Pyrimidine and Glutamate metabolisms, known to play a regulatory role in bone metabolism. Obesity and estrogen deficiency exert combined deleterious effects on bone resulting in accelerated cellular senescence, expansion of BMAT and impaired bone formation leading to decreased bone mass. Our results suggest that obesity may increase bone fragility in post-menopausal women.

Project description:The aim of the current study was to characterize the differential cellular and exosomal miRNAs during inflammation or high fat diet-induced obesity in mice. Mesenteric adipose tissue (MAT) and abdominal aorta (AA) from mice fed a normal chow diet (NCD) or a high fat diet (HFD) were harvested for miRNA profiling. MAT-derived adipocytes (MAT-Ad) challenged with either lipopolysaccharide (LPS, 1 µg/ml) or PBS were harvested for miRNA profiling. Meanwhile, miRNAs encapsulated in MAT-Ad-derived exosomes (MAT-Ad-EX) were also analyzed. Hierarchical clustering analysis performed on most significantly regulated miRNAs (HFD vs NCD in tissues; LPS challenge vs PBS in the cells) showed a set of miRNAs that are differentially expressed in obese versus lean MAT or AA tissues, and in LPS-challenged versus PBS-treated MAT-Ads. The dysregulated of miRNAs in MAT-Ad-EX was also generated and hierarchically clustered, induced by prolonged exposure to microbial product.

Project description:Exosomes carry cell-specific proteins or mRNAs/miRNAs that mediate the functional effect of exosomes. To investigate whether miRNAs within GATA4-Exo are important for their cardioprotective effects, we utilized an unbiased approach by performing an exosomal miRNA array analysis. Total RNA was extracted from CDCs-Exo by using a Qiagen miRNeasy Mini Kit. Global expression patterns of miRNAs were then examined by using Affymetrix GeneChip miRNA 4.0 arrays.Expression data were generated by using Affymetrix Expression Console software and normalized according to the MAS5 method. The RVM t test was applied to filter the differentially expressed genes for the control and experimental groups. Differentially expressed genes were defined based on a P-value threshold and fold-change analysis.
