Project description:We identified 5 distinct fibroblast subsets by sequencing 74957 single cells derived from 7 HBV-related HCC tissues, of which the CD36+ CAFs were highlighted, with enriched lipid metabolism and secreting high levels of macrophage migration inhibitory factor (MIF). CD36+ CAFs had tight interactions with CD33+ MDSCs through MIF/CD74 axis.

HBV remodels PP2A complexes to rewire kinase signaling in hepatocellular carcinoma

Project description:Hepatitis B virus (HBV) infection promotes liver cancer initiation by inducing inflammation and cellular stress, but its impact in established tumors is not well understood. We used affinity purification mass spectrometry to comprehensively map a network of 145 physical interactions between HBV and human proteins in hepatocellular carcinoma (HCC). A subset of the host factors targeted by HBV proteins are preferentially mutated in non-HB¬V-associated HCC suggesting that their interaction with HBV influences HCC biology. These include proteins involved in mRNA splicing, mitogenic signaling and DNA repair, with the latter set interacting with the HBV oncoprotein X (HBx). We show that HBx remodels the PP2A phosphatase complex by excluding striatin regulatory subunits from the PP2A holoenzyme. We find that HBx effects on PP2A cause Hippo kinase activation, particularly in cells with high striatin levels. In parallel, HBx activates mTOR complex 2 (mTORC2) to prevent YAP degradation. mTORC2 effects on YAP can be observed in human HCC specimens and mouse HCC models and can be targeted with mTOR kinase inhibitors. Thus, HBV interaction with host proteins rewires HCC signaling rather than directly activating mitogenic pathways. These findings provide a new paradigm for the cellular effects of a tumor promoting virus and support a model where HBV may have therapeutic actionable effects on HCC biology.

MicroRNA profile differences in Hepatitis B virus (HBV) positive and negative hepatocellular carcinoma

Project description:MicroRNAs (miRNAs) exhibit essential regulatory functions related to cell growth, apoptosis, development and differentiation. Dysregulated expression of miRNAs is associated with a wide variety of human diseases. As such miRNA signatures are valuable as biomarkers for disease and for making treatment decisions. Hepatitis B virus (HBV) is a major risk factor for hepatocellular carcinoma (HCC). Here we screened for miRNAs in chronic HBV associated HCC. To evaluate the effect of HBV infection on the change in expression of miRNAs, 12 pairs of samples from HCC and non-tumor tissues (including 6 HBV-positive HCC and 6 HBV-negative HCC and their non-tumor tissues) were collected. The extracted RNAs were evaluated to detect the expression of miRNAs. Using ANOVA to screen the differential expression of miRNAs at P-value â¤ 0.01, fold change â¥ 2 or â¤ 0.5, 225 miRNAs were detected.

The fibroblast-centered cellular crosstalk in HBV-positive liver cancer revealed by single cell sequencing

Project description:Here, we generated full-length single cell transcriptome data for ~9,000 cells from clinical samples of 11 liver cancer patients to delineate cellular relationships of diverse malignant, immune and stromal cells. We identified an immunomodulatory fibroblast population, FAP+CCL19+CCL21+ CAFs, that expressed signatures of fibroblastic reticular cells and was associated with favorable prognosis in hepatocellular carcinoma (HCC). These CAFs exhibited the potential to recruit NK cells and regulate tumor-associated macrophages, and appeared to be regulated by CXCL12+ endothelial cells in tumors through specific ligand-receptor interaction. Further, we uncovered clonal HBV integration events enriched in tumor cells, and trajectory analysis on tumor cells with clonal HBV integration events revealed that hepatoblast-like cells differentiated into a terminal state with high expression of an immune-resistance program. The intercellular crosstalks between CAFs and cancer cells, macrophages, and NK cells, combined with HBV-based lineage tracing, provide a better understanding for functional roles of CAFs in the HCC microenvironment, and the relationship between HBV integration and liver malignancies.

Project description:Hepatitis B virus (HBV) infection could cause hepatitis, liver cirrhosis and hepatocellular carcinoma. HBV-mediated pathogenesis is only partially understood, but X protein (HBx) reportedly possesses oncogenic potential. Exosomes are small membrane vesicles with diverse functions released by various cells including hepatocytes, and HBV harnesses cellular exosome biogenesis and export machineries for virion morphogenesis and secretion. Therefore, HBV infection might cause changes in exosome contents with functional implications for both virus and host. In this project, exosome protein content changes induced by HBV and HBx were quantitatively analyzed by SILAC/LC-MS/MS. Exosomes prepared from SILAC-labeled hepatoma cell line Huh-7 transfected with HBx, wildtype or HBx-null HBV replicon plasmids were analyzed by LC-MS/MS.

Next Generation Sequencing Identification of HBV-MLL4 integration and its molecular basis in Chinese hepatocellular carcinoma

Project description:Purpose: To gain molecular insights of HBV integration that may contribute to HCC tumorigenesis, we performed whole transcriptome sequencing and whole genome copy number profiling of hepatocellular carcinoma (HCC) samples from 50 Chinese patients. Results: We identified a total of 33 HBV-human integration sites in 16 of 44 HBV-positive HCC tissues, which were enriched in HBV genotype C-infected patients. In addition, significantly recurrent HBV-MLL4 integration (18%; 8/44) was found in this cohort of patients. Conclusions: This is the first report on the molecular basis of the MLL4 integration driving MLL4 over-expression. HBV-MLL4 integration occurred frequently in Chinese HCC patients, representing a unique molecular segment for HCC with HBV infection.
