Project description:Mice carrying a mutation which deletes Bmal1 in Gfap-expressing astrocytes, and control animals were fed either a standard diet or a high fat diet for 16 weeks. Conditional deletion of Bmal1 in Gfap cells decreased weight gain and increased energy expenditure under high fat diet, hence the transcriptome of Brown Adipose Tissue (BAT) and Ventromedial Hypothalamus (VMH) were obtained.

The Circadian Clock in the Ventromedial Hypothalamus Controls Cyclic Energy Expenditure.

Project description:Organismal homeostasis relies on coherent interactions among tissues, specifically between brain-driven functions and peripheral metabolic organs. Hypothalamic circuits compute metabolic information to optimize energetic resources, but the role of the circadian clock in these pathways remains unclear. We have generated mice with targeted ablation of the core-clock gene Bmal1 within Sf1-neurons of the ventromedial hypothalamus (VMH). While this mutation does not affect the central clock in the suprachiasmatic nucleus (SCN), the VMH clock controls cyclic thermogenesis in brown adipose tissue (BAT), a tissue that governs energy balance by dissipating chemical energy as heat. VMH-driven control is exerted through increased adrenergic signaling within the sympathetic nervous system, without affecting the BAT's endogenous clock. Moreover, we show that the VMH circadian clock computes light and feeding inputs to modulate basal energy expenditure. Thus, we reveal a previously unsuspected circuit where an SCN-independent, hypothalamic circadian clock controls BAT function, energy expenditure, and thermogenesis.

Project description:Microdissected ventromedial hypothalamus (VMH) was profiled to identify transcriptional changes after Nkx2-1 ablation in female mice. Total RNA was extracted from conditional mutants (Nkx2-1 f/f; Sf1Cre) and floxed controls (Nkx2-1 f/f) at postnatal day 10 (P10) and profiled using Mouse ref8 v2.0 Illumina chips and reagents (n=4/group). The tissue profiled is microdissected ventromedial hypothalamus from P10 female mice. Controls and mutants are analyzed in quadruplicate.

Project description:The ventrolateral subdivision of the ventromedial hypothalamus (VMHvl) is essential for innate social behaviors including aggression and mating in female mice at different reproductive states. However, the female VMHvl transcriptomes in different reproducive states and their relationship behavioral functions are unknown. We performed single-cell RNA 10x sequencing and investigated correspondence between transcriptomic identity in different reproductive states and behavioral activation. Immediate early gene analysis from our Act-seq dataset identified that distinct T-types function in female aggression and mating behaviors.

Estrogen receptor alpha signaling establishes a sexually dimorphic regulatory node of energy expenditure

Project description:Estrogen receptor a (ERa) signaling in the ventromedial hypothalamus (VMH) contributes to energy homeostasis by modulating physical activity and thermogenesis. However, the precise neuronal populations involved remain undefined. Using single-cell RNA transcriptomics and in situ hybridization, we describe six neuronal populations in the mouse VMH. ERa is enriched in populations showing sex biased expression of reprimo (Rprm), tachykinin 1 (Tac1), and prodynorphin (Pdyn). Female biased expression of Tac1 and Rprm is patterned by ERa-dependent repression during male development, whereas male biased expression of Pdyn is maintained by circulating testicular hormone in adulthood. Chemogenetic activation of ERa+ VMH neurons stimulates heat generation and movement in both sexes. However, silencing Rprm gene function increases core temperature selectively in females and ectopic Rprm expression in males is associated with reduced core temperature. Together these findings reveal a role for Rprm in temperature regulation and ERa in the masculinization of neuron populations that underlie energy expenditure.

Thyroid hormone receptor beta in the ventromedial hypothalamus is essential for the physiological regulation of food intake and body weight

Project description:The obesity epidemic is a significant global health issue. Improved understanding of the mechanisms which regulate appetite and body weight will provide the rationale for the design of novel anti-obesity therapies. Thyroid hormones play a key role in metabolic homeostasis through their interaction with thyroid hormone receptors (TR) which function as ligand inducible transcription factors. The TR beta isoform (TRβ) is expressed in the ventromedial hypothalamus (VMH), a brain area important for control of energy homeostasis. Here we report that selective knock down of TRβ in the VMH of adult mice results in severe obesity due to hyperphagia and reduced energy expenditure. The observed increase in body weight is of a similar magnitude to murine models of the most extreme forms of monogenic obesity. These data identify TRβ in the VMH as a major physiological regulator of food intake and energy homeostasis.
