Project description:Autologous skin flap transplantation is a common method to repair complex soft tissue defects caused by cancer, trauma, and congenital malformations, among other conditions; however, limited blood supply range and post-transplantation ischemia-reperfusion injury (IRI) can cause distal necrosis of the flap and loss of long-term function, severely limiting the indications for this type of surgery. Here, development of a bio-multifunctional hydrogel for programmatic co-delivery medicine, DCDM, that meets the blood supply requirements of skin flap transplantation and can deliver two types of gas molecules, CO and NO, in a timely manner and with a concentration gradient during surgery, is described. DCDM maintains the immediate opening of blood flow channels in transplanted tissue and effective blood perfusion throughout the perioperative period, activating perfusion of the hemodynamic donor site. These results demonstrate that DCDM promotes distal vascularization and long-term functional reconstruction of transplanted tissues by inhibiting inflammatory damage and accelerating blood vessel formation. The findings presented here are applicable to the extended fields of large soft tissue defect repair and transplant of various organs.

Project description:Following severe spinal cord injury (SCI), dysregulated neuroinflammation causes neuronal and glial apoptosis, resulting in scar and cystic cavity formation during wound healing process and ultimately atrophic nerve regrowth inhibitory microenvironment. Because of this complex and dynamic nature of SCI pathophysiology, a systemic solution for scar and cavity free wound healing while remodeling microenvironment for nerve regrowth has been rarely explored. To address this challenge, we provided a one-step solution through a self-assembly, multifunctional hydrogel depot punctually releasing the anti-inflammatory drug methylprednisolone sodium succinate (MPSS) and growth factors locally according to its pathophysiology to repair severe SCI. We found release of growth factors alone could only provide some tissues/axons protection, but failed to eliminate all cystic cavity formation. Sustained release of MPSS is sufficient to modulate dysregulated neuroinflammation and eliminate almost all cystic cavity but unable to alleviate impenetrable scar boundaries, which are largely responsive for nerve regrowth failure. Strikingly, synergically releasing anti-inflammatory drugs MPSS and growth factors in the hydrogel depot along SCI pathophysiology protected spared tissues/axons from secondary injury, promoted scar boundary and cavity free wound healing and made permissive bridges for remarkable axonal regrowth. Behavior and electrophysiology studies indicated that the remnant of spared axons but not regenerating axons mediated functional recovery, strongly suggesting that additional interventions are still required to make the rebuilt neuronal circuits perform functions. Taken together, these findings pave the way to develop a systemic solution to treat acute SCI.

Project description:The native microenvironment following osteochondral defect is not permissive for its endogenous regeneration. A variety of hydrogels have been developed to regulate cell behaviors; however, there is still a lack of hydrogels with multifunctional properties that can provide a supportive microenvironment for endogenous regeneration of osteochondral defects. In this study, a multifunctional polyphenol-based silk hydrogel was developed and its effectiveness on in situ osteochondral regeneration was investigated. Silk fibroin (SF) was crosslinked by tannic acid (TA) to form a hydrogen-bonded supramolecular network, followed by enzymatic crosslinking using horseradish peroxidase (HRP)/H2O2 to form a stable SF-TA hydrogel. In vitro investigations showed that the proposed SF-TA hydrogel exhibited a multitude of biological effects including scavenging reactive oxygen species (ROS), maintaining cell viability, and promoting the proliferation of bone marrow mesenchymal stem cells (BMSCs), as a result of the incorporation of the bioactive TA molecule. Besides, TA had multiple phenolic hydroxyl groups that formed interactions with the therapeutic molecule E7 peptide which improved BMSCs migration when continuously released from the SF-TA hydrogel. In vivo, after implantation for 12-weeks into a rabbit osteochondral defect, the SF-TA hydrogels promoted enhanced regeneration of both cartilage and subchondral bone as compared to hydrogels without TA incorporation. These findings indicated that the multifunctional SF-TA hydrogel provided a microenvironment suitable for the endogenous regeneration of osteochondral defects, thus the SF-TA hydrogel exhibited promising properties for future clinical applications.

To address this issue, we develop a hydrogel patch that releases carbon monoxide and nitric oxide gases on demand, to afford a timely blood supply for skin flap transplantation during surgery. Using an ischemia-reperfusion dorsal skin flap model in rats, we show that the hydrogel patch maintains the immediate opening of blood flow channels in transplanted tissue and effective blood perfusion throughout the perioperative period, activating perfusion of the hemodynamic donor site. We demonstrate that the hydrogel patch promotes distal vascularization and long-term functional reconstruction of transplanted tissues by inhibiting inflammatory damage and accelerating blood vessel formation.

Project description:Diabetic infectious wounds treatment is challenging due to insistent wound infections. Treating such complicated pathological diabetic infectious wounds needs to develop multifunctional materials and understand their mechanism. Here, we developed a novel material termed AgNCs-hydrogel, which is a multifunctional DNA hydrogel which dressing by integrating antibacterial silver nanoclusters. AgNCs-hydrogel was used for promoting the regeneration of diabetic infectious wounds in mouses because that it exhibited superior antibacterial activity, satisfactory biocompatibility, and effective ROS-scavenging property. Based on the skin proteomics, we explored the potential mechanism of AgNCs-hydrogel in treating mouse skin wounds. We found that AgNCs-hydrogel can accelerate proliferation and extracellular matrix (ECM) formation. In proteomic level, AgNCs-hydrogel can regulate some key proteins which mainly located in the extracellular exosome, involved in negative regulation of apoptotic process, performed ATP binding for accelerating diabetic infected wound closure. Therefore, this study provided a multifunctional material AgNCs-hydrogel and revealed its potential mechanisms in promoting the regeneration of diabetic infectious wounds.

Project description:DNA barcodes can be used to identify single cells in a sequencing data space while optical codes can be used to track single live cells in an image data space. We have developed dual image and DNA (ID)-coding, which identifies individual single cells in both live image and sequencing data spaces. Samples provided here are relevant to proof-of-concept studies of ID-coding presented in the associated publication. DNA barcoded micro-particles were encapsulated in hydrogel droplets with or without single cells. The hydrogel droplets were then subjected to “single-droplet sequencing” where whole polyA-bearing nucleic acid components within a hydrogel droplet (i.e. mRNA from cells and synthetic DNA on beads) were concatenated by the same cell barcodes.

Project description:Regenerative medicine aims to tackle a panoply of challenges from repairing focal damage to articular cartilage to preventing pathological tissue remodeling after myocardial infarction. Hydrogels are water-swollen networks formed from synthetic or naturally derived polymers and are emerging as important tools to address these challenges. Recent advances in hydrogel chemistries are enabling researchers to create hydrogels that can act as 3D ex vivo tissue models, allowing them to explore fundamental questions in cell biology by replicating tissues' dynamic and nonlinear physical properties. Enabled by cutting edge techniques such as 3D bioprinting, cell-laden hydrogels are also being developed with highly controlled tissue-specific architectures, vasculature, and biological functions that together can direct tissue repair. Moreover, advanced in situ forming and acellular hydrogels are increasingly finding use as delivery vehicles for bioactive compounds and in mediating host cell response. Here, advances in the design and fabrication of hydrogels for regenerative medicine are reviewed. It is also addressed how controlled chemistries are allowing for precise engineering of spatial and time-dependent properties in hydrogels with a look to how these materials will eventually translate to clinical applications.

Project description:Purpose: To identify genes and the molecular pathways involved in the MSCs response to extracellular matrix stiffness, we performed RNA-sequencing of MSCs which cultured in soft (2 kPa) and stiff (18 kPa) SA hydrogels. Methods: mRNA profiles of MSCs cultured in soft (2 kPa) and stiff (18 kPa) SA hydrogel for 48 h were generated by deep sequencing, in quadruplicate, using Illumina HiSeq 2000. Results: Using an optimized data analysis workflow, we identified 33950 transcripts in MSCs with BWA workflow. Conclusions: Our results present the detailed analysis of MSCs transcriptomes cultured in soft (2 kPa) and stiff (18 kPa) matrix, and found that matrix stiffness dominated multiple mRNA pathways in MSCs.
