Project description:The mononuclear phagocytic system is integral to the clearance of nanotherapeutics. However, the precise mechanisms involved in the phagocytic clearance of nanotherapeutics, and how age-associated changes in innate immune cells affect this clearance, are poorly understood. We found that the ability of professional phagocytes to take up nanoparticles diminishes with age. Bulk RNA sequencing of bone marrow derived macrophages and single cell RNA sequencing of liver macrophages derived from young and old mice was carried out to investigate the gene expression changes that may be assiciated with the changed ability of macrophages to uptake nanoparticles.

Project description:This SuperSeries is composed of the SubSeries listed below.Nanomedicines have been approved to treat multiple human diseases. However, clinical adoption of nanoformulated agents is often hindered by concerns about hepatic uptake and clearance, a process that is not fully understood. Here we show that the antitumour efficacy of cancer nanomedicine exhibits an age-associated disparity. Tumour delivery and treatment outcomes are superior in old versus young mice, probably due to an age-related decline in the ability of hepatic phagocytes to take up and remove nanoparticles. Transcriptomic- and protein-level analysis at the single-cell and bulk levels reveals an age-associated decrease in the numbers of hepatic macrophages that express the scavenger receptor MARCO in mice, non-human primates and humans. Therapeutic blockade of MARCO is shown to decrease the phagocytic uptake of nanoparticles and improve the antitumour effect of clinically approved cancer nanotherapeutics in young but not aged mice. Together, these results reveal an age-associated disparity in the phagocytic clearance of nanotherapeutics that affects their antitumour response, thus providing a strong rationale for an age-appropriate approach to cancer nanomedicine.

TMT isobaric profiling of young versus old haematopoietic stem cells

Project description:Mass spectrometry was performed with an Orbitrap Fusion Tribrid mass spectrometer (Thermo Scientific) interfaced with an UltiMate 3000 Binary RSLCnano System (Dionex). Proteome Discoverer v.1.4 (Thermo Scientific) with SEQUEST HT search engines was used for the spectra-preprocessing and HCD MS2 spectra were used for peptide identification and quantitation based on TMT reporter ions. TMT isobaric comparison of old versus young haematopoietic stem and progenitor cells. Young 1 and Young 2 are samples 126 and 128 of dataset UTH_1. Old 1 and Old 2 are samples 129 and 130 of UTH_1. Young 3 is sample 131 and Old 3 is sample 130 of dataset UTH_4.

High-throughput sequencing for analysis of macropahges from old and young mice.

Project description:Immune cells are essential for proper fracture healing. The goal of this study was to compare the transcriptional profiles of the CD45+ immune cells isolated from the healing fracture callus of old (24 month) and young (3 month) mice (C57BL/6). Analysis of all CD45+ cells was completed via single cell RNAseq. Additional analysis of the macrophages involved in fracture healing in old and young mice was performed via bulk RNAseq. We show that there were multiple subpopulations of macrophages that were transcriptionally heterogenous and present simultaneously within the fracture callus tissue. Attenuated healing in the old mice was associated with a transcriptional profile that shifted towards more pro-inflammatory phenotypes.

Transcriptome analysis of young and old zebrafish ventricles

Project description:The gradual decline of tissue functionality is the main reason why humans suffer from age-related diseases. The prevalence for cardiovascular diseases increases with increasing age. In order to prevent age-related cardiac diseases, it is of importance to understand the respective age-associated risk factors. We have therefore compared the ventricular transcriptome of old and young hearts of the model organism zebrafish. We identified the immune system as activated in the old and found muscle organization to deteriorate upon aging. We show an accumulation of immune cells, mostly macrophages, in the old zebrafish ventricle.
