Project description:Murine colorectal cancer MC38 cells were injected in the flank of C57BL/6 mice to form tumor grafts over 7 days. Tumor-bearing mice were treated with daily injections of vanoxerine (25mg/kg) or vehichle control (saline) over 10 consecutive days. Tumors were excised and RNA sequencing profiling was performed on both experimental condition.

TMT-proteomics of sorted MC38+ tumor cells with high-fat diet

Project description:Obesity is a major cancer risk factor, but the underlying molecular mechanisms are not always known. In this study, we look at proteome remodeling in cancer cells with obesity, comparing tumor cells sorted from mice fed high-fat versus control diet. We conducted 10-plex TMT-proteomics on GFP+ MC38 colorectal adenocarcinoma tumor cells, sorted from subcutaneously implanted tumors 12 days after implantation. This study reveals molecular pathways that change in cancer cells with obesity that promote tumor growth.

RNA Sequencing Analysis of MC38 tumors with Glutamate decarboxylase 1 Knockdown

Project description:Various types of cancers, including colorectal cancer, have shown an unusual dependence on glutamine. Glutamine can undergo an alternate fate in neurons: conversion into a non-proteinogenic amino acid gamma-aminobutyric acid (GABA) by the glutamic acid decarboxylase (GAD), including two family members GAD1 and GAD2. Some studies have shown that GAD1 expression is dysregulated in certain cancer types, but the effect of GAD1 on the tumorigenic process remain largely unknown. Thus, we use mouse colon adenocarcinoma MC38 tumor samples to perform RNA-sequencing (RNA-seq) assay. The goal of this study is to investigate GAD1-mediated transcriptome changes within tumors.

Project description:We show that endothelial cells from MC38 tumors in WT mice express high chemokine Ccl8 than in Apelin knockout mice. We found that Apelin induces Ccl8 expression in ECs and enhances anti-tumor immunity.
