Project description:Background: The initial formation of follicular antrum (iFFA) is a milestone in folliculogenesis, which serves as a boundary between gonadotropin (GTH) -independent folliculogenesis and GTH-dependent folliculogenesis and endows the follicle's ability to acquire response to GTHs and continue to survive. However, iFFA is a barren field in folliculogenesis research, which do not match its physiological significance. Methods:In this study, transcriptome sequencing was used to comprehensively analyze the changes in the life activities of cells in the ovary during iFFA, and then the regulatory mechanism of iFFA was deduced based on the results of omics analysis, and it was systematically verified by experiments. Result: Herein, we conducted in vivo and in vitro experiments to studied iFFA in a mouse model, and revealed that: (1) iFFA is a physiological process of active cell proliferation, increased energy expenditure, enhanced secretory activity and increased moisture absorption, accompanied by complex changes in epigenetic modification, bioclock regulation, and cell junctions; (2) iFFA and blastocoel formation may be conservatively regulated in fluid accumulation, and tight junctions, ion pumps and aquaporins underlie the structural, kinetic and transport foundations for fluid accumulation during iFAA, respectively; (3) mTOR-CNP is the main signal cascade regulating iFFA, it initiated iFFA by targeting tight junction/ion pumps/aquaporins and follicular cell proliferation; (4) FSH controls iFFA by targeting the intra-ovarian mTOR-CNP cascade; (5) mTOR activator could be used as an improver to increase the efficiency of in vitro follicular culture technology. Conclusion: Overall, this study elucidated the changes in follicular cells' life activities in preparation for iFFA, proposed the model of "Dam-Pump-Pipe" to explain the accumulation of follicular fluid, and confirmed that FSH-mTOR-CNP-Tight junction/Ion pumps/Aquaporins is the core cascade controlling iFFA. These findings would help to fill in the gaps in iFFA research, and enrich the theory of folliculogenesis regulation.

Project description:The initial formation of the follicular antrum (iFFA) serves as a dividing line between gonadotropin-independent and gonadotropin-dependent folliculogenesis, enabling the follicle to sensitively respond to gonadotropins for its further development. However, the mechanism underlying iFFA remains elusive. Herein, we reported that iFFA is characterized by enhanced fluid absorption, energy consumption, secretion, and proliferation and shares a regulatory mechanism with blastula cavity formation. By use of bioinformatics analysis, follicular culture, RNA interference, and other techniques, we further demonstrated that the tight junction, ion pumps, and aquaporins are essential for follicular fluid accumulation during iFFA, as a deficiency of any one of these negatively impacts fluid accumulation and antrum formation. The intraovarian mammalian target of rapamycin-C-type natriuretic peptide pathway, activated by follicle-stimulating hormone, initiated iFFA by activating tight junction, ion pumps, and aquaporins. Building on this, we promoted iFFA by transiently activating mammalian target of rapamycin in cultured follicles and significantly increased oocyte yield. These findings represent a significant advancement in iFFA research, further enhancing our understanding of folliculogenesis in mammals.

Project description:Aquaporin-11 (AQP11), a new member of the aquaporin family, is localized in the endoplasmic reticulum (ER). Aqp11–/– mice neonatally suffer from polycystic kidneys derived from the proximal tubule. Its onset is proceeded by the vacuolization of ER. However, the mechanism for the formation of vacuoles and the development of cysts remain to be clarified. Project description:To unravel the genes that underpin the effects of FSH on spermatogenesis, testicular gene expression was compared in juvenile rats after FSH suppression to littermate controls. Based on existing data, it is apparent that the dominant function of FSH shifts between 9 and 18 day postpartum (dpp) during the first wave of spermatogenesis from driving Sertoli cell proliferation to support of germ cells. To enable comprehensive analysis of the impact of acute in vivo FSH suppression on Sertoli and germ cell development and to determine the genes that underpin these affects, FSH was selectively suppressed in Sprague Dawley rats by passive immunisation for 4 days prior to testis collection at 18 days postpartum. FSH suppression did not affect Sertoli cell proliferation at 18 dpp, but germ cell numbers were decreased at 18 dpp following FSH suppression, with a corresponding increase in germ cell apoptosis measured. Sixty transcripts were measured as changed at 18 dpp in response to 4 days of FSH suppression, as assessed using Affymetrix™ microarrays. Some of these are known as Sertoli cell-derived FSH-responsive genes (e.g. StAR, cathepsin L, insulin-like growth factor binding protein-3), while others encode proteins involved in cell cycle and survival regulation (e.g. cyclin D1, scavenger receptor class B 1). These data demonstrate that FSH affects germ cells in vivo, by supporting germ cell viability at day 18. This model enabled identification of candidate genes that contribute to the FSH-mediated pathway by which Sertoli cells support germ cells. Keywords: time course
